摘  要:紫草素是从紫草根部提取的一种天然萘醌类化合物,具有广谱性抗肿瘤作用。本实验就紫草素对人肺癌细胞H292的活性、粘附、迁移及侵袭的影响进行了研究。在使用不同浓度的紫草素工作液处理人非小细胞肺癌H292细胞一定时间后,用MTT法、细胞粘附实验、伤口愈合实验和Transwell实验分别检测细胞活性、粘附、迁移和侵袭能力,并用RT-PCR检测CD54和整合素家族的转录水平、流式细胞术检测CD54蛋白表达水平、Western Blot检测整合素家族蛋白表达水平。实验结果表明:紫草素能以剂量依赖方式抑制H292细胞活性、粘附、迁移和侵袭,并抑制CD54和整合素β1、β3、β4、αv的表达。该研究表明紫草素能通过抑制CD54和整合素家族的表达水平,从而抑制肺癌的转移能力,为肺癌的临床治疗奠定理论基础。As a kind of natural naphthoquinone compound extracted from the root of Lithospermum erythrorhizon. Shikonin has broad spectrum anti-tumor effect. In this paper,the effects of Shikonin on the activity,adhesion,migration and invasion of human lung cancer H292 cells are studied. After human non-small cell lung cancer H292 cells have been treated by Shikonin of different concentrations for a certain period of time,its cell viability and abilities of adhesion,migration and invasion are respectively detected by MTT method,cell adhesion experiment,wound healing and Transwell experiment. The transcription levels of CD54 and integrins family are determined by RT-PCR. The protein expression level of CD54 is detected by flow cytometry while that of integrins family is detected by western blot. The results show that Shikonin could inhibit the activity,adhesion,migration and invasion of H292 cells as well as the expression of CD54,integrin β1,integrinβ3,integrinβ4 and integrinαv in a dose-dependent manner. This study suggests that Shikonin is capable of inhibiting metastasis in lung cancer by inhibiting the expression levels of CD54 and integrins. It has laid a theoretical foundation for the clinical treatment of lung cancer.
