摘  要:在肿瘤微环境(TME)中,肿瘤细胞生长归因于通过各种免疫逃逸机制躲避免疫监视作用,继而形成免疫抵抗。免疫治疗抵抗包括:内因,如肿瘤细胞免疫调节分子的表达改变等;外因,如免疫抑制细胞、可溶性免疫抑制分子或免疫细胞表达的抑制性受体等,将改变肿瘤浸润淋巴细胞的组成和活性。免疫细胞和非免疫细胞之间的交互对话产生正反馈环抑制免疫。虽然免疫治疗已经在不同类型的实体瘤中显示出部分治疗效果,但仍有大部分患者由于获得性抵抗而疗效不佳。这可能由于TME内的细胞和分子抑制网络驱动共同形成免疫抑制微环境。In the tumor microenvironment(TME),tumor cell growth is attributed to evading immune surveillance through various immune escape mechanisms and then forming immune resistence.Immunotherapy resistance includes:the internal factors,such as changes of expression of the tumor cell immune regulatory molecules;extrinsic factors,such as immunosuppressive cells,soluble immunosuppressive molecules or inhibitory receptors expressed by immune cells,which will alter the composition and activity of tumor infiltrating lymphocytes.The interactive dialogue between immune cells and non-immune cells produces a positive feedback loop that enhances immunosuppression.Although immunotherapy has shown therapeutic effects in different types of solid tumors,most of the patients had poor efficacy due to acquired resistance.This may be due to the cellular and molecular inhibition network driving within the TME to form an immunosuppressive microenvironment.
