摘　　要：目的探讨N-乙酰氨基葡萄糖转移酶Ⅴ和Ⅲ(GnT-Ⅴ和GnT-Ⅲ)在胰腺癌中的表达与胰腺癌侵袭和转移的关系。方法采用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶(SP)法、蛋白质印迹法(Western blot)和实时定量反转录聚合酶链反应(RT-qPCR)的方法检测2001年至2007年在天津市南开医院、天津市肿瘤医院手术切除的83例胰腺癌标本和10例正常胰腺组织中GnT-Ⅴ和GnT-Ⅲ基因的表达,并结合临床资料进行分析。采用两组独立样本t检验或单因素方差分析法分析;非正态分布资料采用Wilcoxon秩和检验。定性资料采用χ2检验。结果免疫组织化学结果显示胰腺癌组织GnT-Ⅴ的阳性表达率明显高于正常胰腺组(P<0.01),而GnT-Ⅲ的阳性表达率则明显低于正常胰腺组(P<0.01),GnT-Ⅴ和GnT-Ⅲ的表达均与淋巴结转移、胰腺癌分期显著相关(P<0.05);Western blot和RT-qPCR结果表明胰腺癌的GnT-Ⅴ的蛋白和mRNA的相对表达量明显高于正常胰腺组(P<0.01),而GnT-Ⅲ的蛋白和mRNA的相对表达量明显低于正常胰腺组(P<0.01)。结论 GnT-Ⅴ和GnT-Ⅲ在胰腺癌的侵袭和转移过程中可能起着重要作用,可作为反映胰腺癌生物学行为的一个参考指标。Objective:The study is to investigate the relationship between the expression of N-acetylglucosaminyltransferaseⅤandⅢ(GnT-Ⅴ,GnT-Ⅲ)and invasive or metastasis of pancreatic carcinoma(PCA).Methods:GnT-Ⅴand GnT-Ⅲgene was detected in 83 PCA and 10 normal pancreatic tissues by Immunohistochemistry,real-time quantitative reverse transcriptase-polymerase chain reaction(RT-qPCR)and Western blotting.Results:The expression of the GnT-Ⅴwas positively related to the lymph node metastasis pathological stage;The expresion of the GnT-Ⅲwas negatively related to the lymph node metastasis pathological stage.The expression of GnT-Ⅴprotein and mRNA in PCA was significantly higher than in normal pancreatic tissues.The expression of GnT-Ⅲprotein and mRNA in PCA was significantly lower than in normal pancreatic tissues.Conclusion:GnT-Ⅴand GnT-Ⅲplays a key role in invasiveness and metastasis of PCA,and GnT-Ⅴmight be a valuable marker in reflecting its behavior of PCA.
