摘 要:目的探讨茶多酚对犬骨骼肌缺血再灌注损伤的保护作用。方法取10只健康比格犬作为空白组,10只健康比格犬处理建立犬骨骼肌缺血再灌注损伤模型(模型组),10只健康比格犬处理建立犬骨骼肌缺血再灌注损伤模型,并给予茶多酚预处理(茶多酚组),染色并观察3组组织炎性细胞浸润、纤维化程度、细胞凋亡水平,比较3组GSH、SOD、MPO、HIS、MDA含量,以及IL-1、IL-6、TNF-α水平。结果与空白对照组比较,模型组的组织炎性细胞浸润、组织纤维化程度、组织的细胞凋亡明显增加,茶多酚处理组组织炎性反应、组织纤维化程度、细胞凋亡程度显著降低。模型组动物组织中GSH、SOD、MPO、HIS、MDA的含量较空白组多,茶多酚处理组动物组织中GSH、SOD、MPO、HIS、MDA含量较空白组多,但较模型组少,差异有统计学意义(P<0.05)。模型组动物组织中IL-1、IL-6、TNF-α的含量较空白组多,茶多酚处理组动物组织中IL-1、IL-6、TNF-α的含量较空白组多,但较模型组少,差异有统计学意义(P<0.05)。结论茶多酚预处理对犬骨骼肌缺血再灌注损伤具有一定的保护作用。Objective To investigate the protective effects of tea polyphenols on skeletal muscle ischemia-reperfusion injury in dogs.Methods Ten healthy Beagle dogs served as blank group,10 healthy beagle dogs were treated to establish skeletal muscle ischemia-reperfusion injury model(model group),and 10 healthy beagle dogs were treated to establish skeletal muscle ischemia-reperfusion injury model and pretreated with tea polyphenols(tea polyphenols group).Stain and observe the infiltration,fibrosis and apoptosis levels of inflammatorg cells in the three groups.The contents of GSH,SOD,MPO,HIS,MDA and the levels of IL-1,IL-6 and TNF-αin the three groups were compared.Results Compared with the blank control group,the tissue inflammatory cell infiltration,tissue fibrosis and apoptosis were significantly increased in the model group,while significantly decreased in the tea polyphenols group.The contents of GSH,SOD,MPO,HIS and MDA were more in the model group than in the blank group,more in the tea polyphenols group than in the blank group,less in the tea polyphenols group than in the model group,the difference was statistically significant(P<0.05).The contents of IL-1,IL-6 and TNF-αwere more in the model group than in the blank group,more in the tea polyphenols group than in the blank group,less in the tea polyphenols group than in the model group,the difference was statistically significant(P<0.05).Conclusion Tea polyphenols preconditioning has a certain protective effect on skeletal muscle ischemia-reperfusion injury in dogs.
