摘 要:目的分析帕金森病患者血清胞外囊泡中差异表达微小RNA(miRNA)及其对疾病进展的影响。方法收集该院85例帕金森病患者的临床资料,根据发病年龄段的不同,分为早发组37例和晚发组48例,另以该院30例健康体检人员为对照组。取血清标本的胞外囊泡,采用miRNA表达谱芯片和实时荧光定量聚合酶链反应进行检测和验证,检测帕金森病患者血清胞外囊泡中靶基因的相对表达量。结果晚发组血清胞外囊泡中miR-500a-5p(0.22±0.04)和miR-451(0.25±0.05)表达量较对照组的miR-500a-5p(1.00±0.05)和miR-451(1.01±0.08)降低(P<0.01);而miR-1180-3p(2.06±0.10)和miR-143-3p(2.13±0.14)表达量较对照组miR-1180-3p(1.03±0.04)和miR-143-3p(1.00±0.06)升高(P<0.01)。相比早发组,晚发组血清胞外囊泡中miR-143-3p(4.25±0.19 vs.2.13±0.14)表达量降低(P<0.01)。经Cytoscape分析发现,Akt1和B淋巴细胞瘤-2(Bcl-2)是miR-143-3p的关键靶基因。相比早发组,晚发组血清胞外囊泡中Bcl-2表达量(0.19±0.05 vs 0.78±0.15)升高(P<0.01)。结论miR-143-3p与其靶基因Bcl-2表达水平的异常改变可能与帕金森病的疾病进展密切相关。Objective To analyze the differential expression of microRNAs(miRNA)in the serum extracellular vesicles of patients with Parkinson’s disease(PD)and its effect on PD progression.Methods The clinical data of 85 PD patients were reviewed.Pa-tients were divided into early onset group(n=37)and late onset group(n=48)according to the age of onset.Thirty healthy individ-uals who underwent physical examination at our hospital were used as controls.The relative expression of target genes in the extracellu-lar vesicles of serum samples from PD patients were measured by miRNA microarray and real-time quantitative PCR.Results miR-500a-5p and miR-451 expression was significantly lower in the late onset group than in the control group(0.22±0.04 and 0.25±0.05 vs 1.00±0.05 and 1.01±0.08,P<0.01).In contrast,miR-1180-3p and miR-143-3p expression was significantly higher in the late onset group than in the control group(2.06±0.10 and 2.13±0.14 vs 1.03±0.04 and 1.00±0.06,P<0.01).In addi-tion,miR-143-3p expression in serum extracellular vesicles was significantly lower in the late onset group than in the early onset group(2.13±0.14 vs 4.25±0.19,P<0.01).Cytoscape analysis revealed that Akt1 and B-cell lymphoma-2(Bcl-2)were the key target genes of miR-143-3p.In line with this,Bcl-2 expression in serum extracellular vesicles was significantly higher in the late onset group than in the early onset group(0.78±0.15 vs 0.19±0.05,P<0.01).Conclusions Abnormal changes in the expression of miR-143-3p and its target gene Bcl-2 may be closely related to the progression of PD.
