摘 要:目的探讨在大鼠慢性高眼压模型中,转化生长因子(TGF-β1)在视网膜神经节细胞(RGCs)的表达及抗凋亡作用。方法将54只正常Wistar雄性大鼠随机分为正常组(正常眼压组)6只(12只眼)、模型组(普通高眼压组)24只(48只眼)及干预组(TGF-β1抗体干预高眼压组)24只(48只眼)。用热凝器烧灼巩膜上静脉建立高眼压模型,玻璃体腔注射TGF-β1抗体50 ng建立TGF-β1抗体干预高眼压组,3组分别于建模后1个月内每组取材,免疫组化SP法检测3组大鼠RGCs中TGF-β1蛋白的表达,Realtime PCR法检测RGCs中TGF-β1 mRNA的相对表达量,采用TUNEL法检测RGCs凋亡率。结果(1)TGF-β1蛋白阳性率:正常组RGC细胞中TGF-β1蛋白阳性率为33.33%,模型组为75.00%,而干预组为4.17%。两两比较,模型组与干预组比较,χ^2=22.301,P=0.000,有统计学意义。而干预组与正常组,正常组与模型组比较,均无统计学意义(P>0.05)。(2)TGF-β1 mRNA相对表达量:在正常组RGC细胞中为1.19±0.09,模型组为6.33±4.81,而干预组为0.20±0.17,正常组与模型组比较(t=2.583,P=0.027),模型组与干预组比较(t=3.119,P=0.011),正常组与干预组比较(t=13.667,P=0.000),差异均具有统计学意义。(3)RGC细胞凋亡率:模型组和干预组均有随时间延长凋亡率增加趋势,不同时间点两组比较,干预组均高于模型组,均具有统计学意义(P<0.05)。结论TGF-β1可能作为一种神经营养因子或者抗凋亡因子,阻止视网膜神经节细胞的凋亡。OBJECTIVE To investigate the expression and anti-apoptosis effect of TGF-β1 in retinal ganglion cells(RGCs)of chronic high intraocular pressure in rat models.METHODS Fifty-four normal male Wistar rats were randomly divided into the normal group(normal intraocular pressure group,12 eyes of 6 rats),the model group(ordinary high intraocular pressure group,48 eyes of 24 rats)and the intervention group(high intraocular pressure intervened by TGF-β1 antibody group,48 eyes of 24 rats);the model was established with hot condenser burning sclera vein,the intervention group was established with vitreous injection of TGF-β1 antibodies 50 ng.The eye samples were taken respectively within 1 month.The immunohistochemical SP method was used to detect the expression of TGF-beta 1 protein in RGCs in three groups of rats.Realtime PCR method was used to detect relative expression of TGF-beta 1 mRNA in RGCs in three groups of rats.Meanwhile,TUNEL method was used to detect the apoptosis rate of RGCs in model group and intervention group.RESULTS(1)The positive rate of TGF-beta 1 protein in RGCs was 33.33%in the normal group,75.00%in the model group and 4.17%in the intervention group.Pairwise comparison:The comparison between model group and the intervention group(χ^2=22.301,P=0.000),showed statistically significant differences.However,there were not statistically significant differences between the normal group and the intervention group,the normal group and the model group.(2)The relative expression of TGF-β1 mRNA in RGCs was 1.19±0.09 in the normal group,6.33±4.81 in the model group and 0.20±0.17 in the intervention group.Pairwise comparison:comparison between the normal group and the model group(t=2.583,P=0.027),the model group and the intervention group(t=3.119,P=0.011),the normal group and the intervention group(t=13.667,P=0.000)all showed statistically significant differences.(3)The apoptosis rate of RGCs of the model group and the intervention group both increased with time.Comparison of results between them at differen
