摘 要:脱氢/还原酶因具有高度的区域和立体选择性,常用于生物催化合成在化学、医药和精细化学品等领域有重要应用价值的手性化合物。介导双酮基还原的脱氢/还原酶是特殊的羰基还原酶,在辅酶NAD(P)H的存在下,可催化二酮类底物得到手性双醇。分析了国内外关于介导双酮基还原的脱氢/还原酶酶偶联和底物偶联法辅酶再生的研究进展,提出了辅酶再生循环系统的理性改造策略。Dehydrogenase/reductase is commonly used in biosynthesis of chiral compounds in the fields of chemistry,medicine and fine chemicals due to its high regioselectivity and stereoselectivity.In the presence of the coenzyme NAD(P)H,dehydrogenases/reductases that can mediate diketone reduction are special carbonyl reductases.In this paper,from the perspective of the coenzyme regeneration,we have analyzed the domestic and international research progress on dehydrogenase/reductase which mediates diketone reduction with enzyme-coupled and substrate-coupled methods.Finally,we proposed a rational transformation strategy for the coenzyme-regeneration cycle system.
