摘 要:目的建立切向流超滤富集法(Pellicon超滤膜第一次富集和PEG第二次富集)用于饮用水中诺如病毒的检测。方法梯度稀释含有NoV GⅡ型诺如病毒的粪便,分别加入纯水中制成模拟水样,同时加入MS2标准品作为过程对照,用Pellicon-PEG富集法浓缩后,再用Real-time PCR检测富集前后的NoV GⅡ病毒和MS2标准品浓度,评估该方法的灵敏度、回收率和抑制指数。结果切向流超滤富集法检测灵敏度为模拟水样10-4 Copies/μL,平均回收率为10%~20%,抑制指数均少于2。结论初步建立大体积水样诺如病毒的富集检测方法,为提高水源性诺如病毒胃肠炎疫情暴发实验室检测能力奠定基础。Objective To develop the Pellicon-PEG methody for concentrating and detecting the norovirus in water.Methods The feces which contained the Nov GⅡnorovirus were added to the pure water gradient.At the same time,MS2 standard was added as process comparison.The norovirus in the water were concentrated by Pellicon–PEG methody,and detected by Real-time PCR to evaluate the detection sensitivity,recovery and inhibition index.Results The detection sensitivities of Pellicon-PEG methody was 10-4 Copies/μL of simulated water samples,the recoveries were from 10%to 20%,and the inhibition indexes were less than 2.Conclusion A method for the enrichment and detection of norovirus in large water samples was established preliminary to improve the ability of laboratory detection of the outbreak of waterborne norovirus gastroenteritis.
