摘 要:目的探讨下调c-Met基因的表达对膀胱癌T24细胞放射增敏作用及相关作用机制。方法选取膀胱癌T24细胞,培养后分为空白组、上调c-Met组和下调c-Met组。对各组细胞增殖、凋亡能力、周期分布、放射敏感性进行检测,RT-PCR检测c-Met mRNA表达,Western blot法检测c-Met、Bcl-2、Bax、caspase3表达。结果上调c-Met组细胞c-Met、c-Met mRNA表达均高于空白组,下调c-Met组c-Met、c-Met mRNA表达均低于空白组及上调c-Met组(P<0.05)。在第24、48、72小时,上调c-Met组细胞增殖率高于空白组,凋亡率低于空白组(P<0.05);下调c-Met组细胞增殖率低于空白组及上调c-Met组,凋亡率高于空白组及上调c-Met组(P<0.05)。上调c-Met组细胞处于G1期的比例低于空白组,处于S、G2期的比例高于空白组(P<0.05);下调c-Met组细胞处于G1期的比例均高于空白组及上调c-Met组,处于S、G2期的比例低于空白组、上调c-Met组(P<0.05)。c-Met上调组细胞D0、N、SF2值均高于对照组(P<0.05);下调c-Met组细胞D0、N、SF2值均低于对照组与c-Met上调组(P<0.05)。上调c-Met组细胞Bcl-2、caspase3表达低于空白组,Bax表达高于空白组(P<0.05);下调c-Met组细胞Bcl-2、caspase3表达均高于对照组及c-Met上调组,Bax表达低于对照组与c-Met上调组(P<0.05)。结论下调c-Met基因表达,能够阻滞细胞周期分布,提升膀胱癌T24细胞放射敏感性,调控Bcl-2、Bax、caspase3表达,抑制膀胱癌T24细胞增殖,促进膀胱癌T24细胞凋亡。Objective To investigate the radiosensitization effect of down regulating the expression of c-met gene on bladder cancer T24 cells and its mechanism.Methods T24 cells were divided into blank group,up-regulated c-met group and down regulated c-met group.Cell proliferation,apoptosis,cycle distribution and radiosensitivity were detected.c-met mRNA expression was detected by RT-PCR and c-Met,Bcl-2,Bax and Caspase-3 expression by Western blot.Results The expression of c-met and c-met mRNA in the up-regulated c-met group was higher than that in the blank group.The expression of c-met and c-met mRNA in the down-regulated c-met group was lower than that in the blank group and the up-regulated c-met group(P<0.05).At 24,48 and 72 hours,the cell proliferation rate of the up-regulated c-met group was higher than that of the blank group.The apoptosis rate of the down-regulated c-met group was lower than that of the blank group and the up-regulated c-met group.The apoptosis rate of the down-regulated c-met group was higher than that of the blank group and the up-regulated c-met group(P<0.05).The proportion of cells in G1 phase in up-regulated c-met group was lower than that in blank group.The proportion in S and G2 phase was higher than that in blank group;the proportion in G1 phase in down-regulated c-met group was higher than that in blank group and up-regulated c-met group.The proportion in S and G2 phase was lower than that in blank group and up-regulated c-met group(P<0.05).The values of d0,N and SF2 in the up-regulated c-met group were higher than those in the control group,and the values of d0,N and SF2 in the down-regulated c-met group were lower than those in the control group and the up-regulated c-met group(P<0.05).The expression of Bcl-2 and Caspase-3 in the up-regulated c-met group was lower than that in the blank group,and the expression of Bax in the down-regulated c-met group was higher than that in the control group and the up-regulated c-met group.The expression of Bax was lower than that in the control group
