摘  要:以Ensifer meliloti LZgn5(苜蓿内源根瘤菌)和E.meliloti 12531(苜蓿外源根瘤菌)作为研究对象,研究硼对内源和外源根瘤菌生长、胞外多糖和生长素(IAA)产量、固氮酶活性以及菌体形态结构的影响。试验共设5个处理,未加硼的菌液为对照组、gn5f和gn5o(100mg/L硼处理荧光蛋白标记和非标记E.meliloti LZgn5)以及12531f和12531o(1mg/L硼处理荧光蛋白标记和非标记E.meliloti 12531f)。结果表明,100mg/L硼处理gn5f和gn5o以及1mg/L硼处理12531f和12531o,均可促进相应根瘤菌的生长、胞外多糖及IAA产量,同时可以改变菌体形态及表面结构,但对固氮酶活性无影响。为硼促进内源和外源根瘤菌在紫花苜蓿体内运移、定殖机理研究提供间接依据。Ensifer meliloti LZgn5(alfalfa endogenous rhizobium)and E.meliloti 12531(alfalfa exogenous rhizobium)were used as research subjects to study the effect of boron on the growth,extracellular polysaccharides and IAA production,nitrogenase activity and morphological structure of endogenous and exogenous rhizobium.Five treatments were set in the present study,including boron-free rhizobia solutions as control,boron treated fluorescent protein-tagged E.meliloti LZgn5f(gn5f)and untagged E.meliloti LZgn5(gn5o)solutions(100 mg/L),boron treated fluorescent protein-tagged E.meliloti 12531f(12531f)and untagged E.meliloti 12531o(12531o)solutions(1mg/L).The results showed that the growth,extracellular polysaccharides and IAA production of endogenous exogenous and rhizobium were promoted by boron treated solution.The cell morphology and surface structure of endogenous exogenous and rhizobium were also changed by boron treated solution.However,the nitrogenase activity of rhizobium among all treatments was not affect by boron addition.This study provided basis for the mechanism of boron promoted migration and colonization of endogenous and exogenous rhizobia in alfalfa.
