摘  要:目的探讨长链非编码RNA HAGLROS(lncRNA HAGLROS)对非小细胞肺癌A549细胞系迁移和侵袭能力的影响.方法采用siRNA(小RNA干扰技术)对细胞进行转染;同时采用实时荧光定量PCR技术(qPCR)检测相关目的基因的表达水平;应用细胞划痕实验检测细胞的迁移能力;应用Transwell实验检测细胞的侵袭能力.结果qPCR结果显示:对A459细胞进行转染后,siRNA-lncRNA HAGLROS-homo-435敲减效率最高,差异具有统计学意义(P<0.05);划痕实验和Transwell实验结果表明:实验组与对照组比较,细胞迁移能力、细胞侵袭能力均下降,差异具有统计学意义(P<0.05).结论lncRNA HAGLROS在肺癌细胞系A549中作为促癌基因发挥作用,可为肺癌的诊断及治疗提供临床参考.Objective To investigate the effect of long non-coding RNA HAGLROS(lncRNA HAGLROS)on migration and invasion of non-small cell lung cancer A549 cell line.Method Cells were transfected with siRNA(small RNA interference technology);the expression level of related target genes was detected by real-time fluorescence quantitative PCR(qPCR);cell scratch test was used to detect cell migration ability;Transwell test was used to determine cell invasion ability.Results The qPCR results showed that siRNA-lncRNA HAGLROS-homo-435 had the highest knock-down efficiency after transfection of A459 cells(P<0.05).Scratch results and Transwell results showed that the migration ability and invasive ability of cells in the experimental group were decreased comparing with the control group,and the difference was statistically significant(P<0.05).Conclusion lncRNA HAGLROS is an oncogene in lung cancer cell line A549,which can provide clinical diagnosis and treatment for lung cancer.
