摘 要:为了探究条石鲷精子最适紫外灭活剂量及其诱导大黄鱼雌核发育的效果,利用Ringer氏液将条石鲷精子稀释40倍后,置于辐射强度1330~1350μW·cm^-2的紫外灯下进行灭活,然后与正常大黄鱼卵子进行人工授精和冷休克处理(水温3℃,受精2 min后,冷休克10 min)。结果表明:条石鲷精子最适紫外灭活剂量为80 mJ·cm^-2,随着照射剂量的增加,条石鲷精子表现出典型的Her-twig效应。利用流式细胞仪检测倍性,未经冷休克处理的受精卵全部为单倍体,冷休克处理的均为二倍体,表明精子灭活有效。性腺组织切片结果显示了雌核发育群体的全雌性。实验结果表明:条石鲷精子可作为一种合适的激活源,诱导大黄鱼的雌核发育。This study investigated the optimal UV irradiation dosage of Oplegnathus fasciatus sperm to induce meiogynogenesis in Larimichthys crocea.Milt from O.fasciatus males was diluted 1:40 with Ringer’s solution and UV irradiated at an intensity of 1330-1350μW·cm^-2,and used to fertilize L.crocea eggs.The fertilized eggs were treated with a cold-shock(3℃for 10 min at 2 min after fertilization).The result showed the optimal UV irradiation of the O.fasciatus sperm for gynogenesis was 80 mJ·cm^-2.A typical“Hertwig effect”was observed based on the fertilization and hatching of haploid by subjecting eggs fertilized with UV-irradiation sperm.Fries of gynogenesis were confirmed to be diploids via flow cytometer,and all the juvenile gynogens examined by histology were female.This study provided an effective method to induce meiogynogenesis in L.crocea.
