摘 要:为研究日本黄姑鱼鱼皮免疫活性肽酶解工艺,以日本黄姑鱼鱼皮为原料,以小鼠单核巨噬细胞RAW264.7的相对增殖率为评价指标,筛选最优酶种并考察pH、温度、时间、加酶量、液料比对小鼠巨噬细胞RAW264.7相对增殖率的影响。再经单因素实验,响应面分析法对其酶解工艺进行优化。结果表明,最优酶种和酶解条件为中性蛋白酶,pH 7.0,温度45.5℃,时间5.5 h,加酶量1500 U·g^-1,液料比10:1(mL·g^-1),所得酶解产物作用于RAW 264.7的相对增殖率为57.47%。本研究为日本黄姑鱼鱼皮的高附加值利用提供依据。In order to prepare immunomodulatory peptides from the skin of Nibea japonica,the enzymatic hydrolysis process was optimized according to the relative proliferation rate of macrophage RAW 264.7.And the single factor and response surface methodology were performed to investigate the effects of extraction parameters on immunomodulatory peptides from the skin of the N.japonica.The optimum extraction conditions of immunomodulatory peptides were as follows:the concentration of Neutral protease was 1500 U·g^-1,pH was 7.0,temperature was 45.5℃,solid-liquid ratio was 1:10(mL·g^-1),and hydrolysis time was 5.5 h.Under these conditions,the relative proliferation rate of the obtained enzymatic hydrolysate to RAW 264.7 was 57.47%.Above all,our study provides a basis for the conversion of by-product(fish skin)that produced in the processing of N.japonica to high value-added products.
