摘 要:目的探讨山药多糖对肿瘤小鼠的抗肿瘤作用和免疫调节作用的研究。方法选取C57BL/6小鼠,Lewis肺癌细胞,随机分成正常对照组(NC)、高剂量组(HD)、中等剂量组(MD)、低剂量组(LD),每组10只,接种0.2 mL,Lewis肺癌肿瘤细胞悬液,LD、MD和HD组每天分别灌胃给予50、150和250 mg/kg的山药多糖一次,NC和BC组给予等量的生理盐水,连续7 d。结果小鼠体内T淋巴细胞增值能力显著降低,从剂量上观察,采用中剂量的疗效最佳,其T淋巴细胞增值能力为(28185±1145)min,均显著高于对照组[(28236±1030)min]、肿瘤小鼠组[(14680±1420)min]、低剂量组[(19226±1038)min]和高剂量组[(27685±1159)min],且调节使具有明显双向性。小鼠体内NK细胞的活性显著降低,给予山药多糖后,低剂量组[(7.6±0.8)%]仍不能恢复至对照组水平[(13.3±1.32)%],而中[(14.5±1.14)%]、高剂量[(12.6±1.45)%]均能达到对照组更高或基本相同水平,可良好增强细胞免疫功能,比较有统计学意义(P<0.05);小鼠经处理后剥离实体肿瘤进行称重计算抑瘤率,表明山药多糖可抑制肿瘤的生长,对肿瘤的抑制率高达64.00%。肿瘤小鼠IL-2[(0.86±0.14)U/mL]和TNF-α水平明显下降(P<0.05),但山药多糖可显著提高肿瘤小鼠IL-2和TNF-α水平,剂量越高,肿瘤小鼠IL-2和TNF-α水平恢复越接近正常。结论山药多糖可有效抑制小鼠体内肿瘤的生长,并具有显著免疫调节的效用。Objective To investigate the anti-tumor and immunomodulatory effects of Yam polysaccharides on tumor mice.Methods C57 BL/6 mice,Lewis cells of lung cancer were selected and randomly divided into normal control group(NC),high dose group(HD),medium dose group(MD),low dose group(LD),10 rats in each group,inoculated with 0.2 mL tumor cell suspension of Lewis lung cancer.The LD,MD,and HD groups were given 50,150,and 250 mg/kg of Yam polysaccharide respectively once a day,and the NC and BC groups were given the same amount of normal saline for 7 days.Results The ability of T lymphocyte proliferation in mice significantly reduced.For the dose,the therapeutic effect of medium dose was the best,and its ability of T lymphocyte proliferation was(28185±1145)min,which was significantly higher than that in the control group(28236±1030)min,the tumor mice group(14680±1420)min,the low dose group(19226±1038)min and the high dose group(27685±1159)min,and the adjustment had obvious bidirectionality.The activity of NK cells in mice significantly reduced.After the mice were given Yam polysaccharides,the level in low-dose group(7.6±0.8)%could not be returned to the level in the control group(13.3±1.32)%,while the level in medium-dose group(14.5±1.14)%and high-dose group(12.6±1.45)%could be higher or almost equal,which could enhance the cellular immune function.The difference was statistically significant(P<0.05).After the mice were treated,the tumors were stripped from entity and weighed to calculate the tumor inhibition rate,which indicated that the Yam polysaccharide could inhibit the growth of the tumor,and the inhibition rate of the tumor was as high as 64.00%.The levels of IL-2(0.86±0.14)(U/mL)and TNF-αin tumor mice significantly decreased(P<0.05),but Yam polysaccharide could significantly increase the levels of IL-2 and TNF-αof tumor mice.The higher the dose was,the more normal the levels of IL-2 and TNF-αof tumor mice were.Conclusion Yam polysaccharide can effectively inhibit the growth of tumors in mice and has sig
