摘 要:目的观察地佐辛对切口痛大鼠中脑导水管周围灰质5-HT7受体表达影响。方法2017年12月至2018年6月由郑州大学动物实验中心提供清洁级成年雄性Wistar大鼠,48只,体重250~300 g,随机分为4组(n=12):C组为对照组仅行麻醉而未行手术;S组为生理盐水组:在大鼠切口痛模型建造前15 min尾静脉注射生理盐水(2 mL);D1组为1 mg/kg地佐辛组:大鼠切口痛模型建造前15 min尾静脉注射地佐辛1 mg/kg(2 mL);D2组为3 mg/kg地佐辛组:切口痛模型建造前15 min尾静脉注射地佐辛3 mg/kg(2 mL)。于术前24 h(T0)、术后2 h、6 h、24 h(T1~3)时测定机械缩足反射阈值(MWT)和累积疼痛评分,T3时痛阈测定后处死大鼠,取大鼠全脑,检测中脑导水管周围灰质神经元5-HT7受体表达。结果与C组比较,S组,D1组和D2组T1~3时MWT明显降低,累积疼痛评分升高,中脑导水管周围灰质神经元5-HT7受体免疫阳性细胞数增高(P<0.05);与S组比较,D1组和D2组T1~3时MWT升高,累积疼痛评分降低,中脑导水管周围灰质神经元5-HT7受体免疫阳性细胞表达增加(P<0.05);与D1组比较,D2组T1-3时MWT升高,累积疼痛评分降低,中脑导水管周围灰质神经元5-HT7受体免疫阳性细胞表达增加(P<0.05)。结论地佐辛可促进中脑导水管周围灰质神经元5-HT7受体表达,减轻大鼠切口痛,且表达水平随剂量增加而增加。Objective To observe the effect of dezocine in periaqueductal gray of incisional pain in rat models.Methods Forty-eight Wistar male rats that weighed 250-300 g were randomly divided into four groups(n=12 each)using a number table:control group(group C),normal saline group(group S),dezocine 1 mg/kg group(group D1)and dezocine 3 mg/kg group(group D2).Rats in the control group were anesthetized without surgery.In the saline or dezocine D1,D2 group,rats received tail intravenous injection of normal saline 2 mL,dezocine 1 mg/kg or dezocine 3 mg/kg(in 2 mL of volume)15 min before getting incisional pain models.The mechanical paw withdrawal threshold(MWT)and cumulative pain score were measured 24 h before surgery(T0,baseline)and 2 h,6 h,24 h after surgery(T1-3).The rats were sacrificed after measurement of pain threshold at T1-3 and the total brains were removed for detection of 5-HT7 receptor expression in the periaqueductal gray(by immune-histochemistry).Results Compared with group C,Significantly,MWT was decreased,cumulative pain score was increased,and the expression of 5-HT7 receptor protein was up-regulated at T1-3 in S,D1 and D2 groups.Compared with group S,MWT was significantly increased,cumulative pain score was decreased,and the expression of 5-HT7 receptor protein was up-regulated at T1-3 in D1,D2 groups.Compared with group D1,MWT was significantly increased,cumulative pain score was decreased,and the expression of 5-HT7 receptor protein was up-regulated at T1-3 in group D2.Conclusion Dezocine can enhance up-regulation of 5-HT7 receptor protein expression and improve the expression level with the increase of the dosage,thus attenuating incisional pain in rats.
