摘  要:目的探讨DNA甲基转移酶1在卵巢癌中的表达意义及其作用机制。方法收集手术切除的31例卵巢癌患者新鲜手术标本,采用免疫组化方法检测15例正常卵巢组织及31例卵巢癌组织中DNMT1的表达水平。结果DNMTl在卵巢癌组织中的表达明显高于正常卵巢组织。临床期别越高、病理级别越高以及伴随远处转移的卵巢癌组织中DNMT1的阳性率越高。在SKOV3细胞中,转染DNMT1 siRNA敲低DNMT1后,表明DNMT1蛋白水平明显下降,CCK8实验结果证实敲低DNMT1后SKOV3的增殖能力下降,Transwell小室实验结果提示敲低DNMT1后SKOV3迁移能力下降。结论DNA甲基转移酶1在卵巢癌组织中的表达阳性率增高,且与卵巢癌临床期别、病理级别以及伴随远处转移有一定的相关性,可促进卵巢癌细胞增殖,影响癌症进展。Objective To investigate the expression of DNA methyltransferase 1 in ovarian cancer and its effect on the prognosis of ovarian cancer.Methods The expression of DNMT1 in 15 normal ovarian tissues and 31 ovarian cancer tissues were detected by immunohistochemistry.Results DNMT1 was a key enzyme to maintain DNA methylation,and its expression was closely related to gene imprinting,cell differentiation and development,and tumor formation.The higher the clinical stage,the higher the pathological grade,and the higher the positive rate of DNMT1 in ovarian cancer tissues with distant metastasis.In SKOV3 cells,the DNMT1 protein level was significantly decreased after DNMT1 siRNA transfection was knocked down,and the CCK8 results confirmed that SKOV3 proliferation ability was decreased after DNMT1 knockdown,and the Transwell chamber results suggested that SKOV3 migration ability was decreased after DNMT1 knockdown.Conclusion The expression of DNMTl in ovarian cancer tissues is significantly higher than that of normal ovarian cancer tissues.
