补康灵方对小鼠Lewis肺癌抑瘤作用及其机制研究

摘  要:目的观察补康灵方对小鼠Lewis肺癌的抑瘤作用及对血管内皮生长因子(VEGF)、细胞周期素D1(Cyclin D1)、凋亡基因蛋白Bcl-2/Bax表达的影响,探讨补康灵汤的抗肿瘤作用机制。方法按体质量将40只成功建立Lewis肺癌模型小鼠随机分为4组,模型组给予生理盐水灌胃,中药组给予补康灵方灌胃,化疗组给予环磷酰胺隔日腹腔注射,中药+化疗组给予补康灵汤灌胃和环磷酰胺隔日腹腔注射,均干预12 d。比较各组小鼠移植瘤的抑瘤率,采用SP免疫组化染色法检测各组小鼠瘤组织中VEGF、Cyclin D1表达情况,采用Western blot法检测各组小鼠瘤组织中Bcl-2、Bax表达情况,计算Bcl-2/Bax比值。结果中药组、化疗组、中药+化疗组抑瘤率分别为24.6%,45.6%,59.8%,中药+化疗组明显高于化疗组和中药组(P均<0.05)。中药组、化疗组、中药+化疗组瘤组织中VEGF、Cyclin D1光密度和Bcl-2表达水平、Bcl-2/Bax比值均明显低于模型组(P均<0.05),Bax表达水平均明显高于模型组(P均<0.05);中药+化疗组瘤组织中VEGF、Cyclin D1光密度和Bcl-2表达水平、Bcl-2/Bax比值均明显低于中药组和化疗组(P均<0.05),Bax表达水平明显高于中药组和化疗组(P均<0.05)。结论补康灵方具有一定的抑瘤作用,其抗肿瘤作用可能与下调VEGF、Cyclin D1、Bcl-2表达,调节Bcl-2/Bax比值,干预细胞周期,促进肿瘤细胞凋亡有关。Objective It is to observe the anti-tumor effect of Bukangling Decoction on Lewis lung cancer in mice and its effect on the expression of vascular endothelial growth factor(VEGF),Cyclin D1(Cyclin D1)and apoptosis gene protein Bcl-2/Bax,and to explore the anti-tumor mechanism of Bukangling Decoction.Methods According to body weight,40 mice successfully established Lewis lung cancer model were randomly divided into 4 groups.The model group was given normal saline by gavage,the Chinese medicine group was given Bukangling Decoction by gavage,and the chemotherapy group was given cyclophosphamide by intraperitoneal injection every other day,Chinese medicine+chemotherapy group(combination group)was given Bukangling Decoction by gavage and cyclophosphamide by intraperitoneal injection every other day,all the groups were treated for 12 days.The tumor inhibition rate of xenograft tumors in each group was compared,the expression of VEGF and Cyclin D1 in the tumor tissue of each group was detected by SP immunohistochemical staining method,and the expression of Bcl-2,Bax in tumor tissue of each group was detected by Western blot,and the Bcl-2/Bax ratio was calculated.Results The tumor inhibition rates of the Chinese medicine group,the chemotherapy group,and the combination group were 24.6%,45.6%,and 59.8%,respectively,the rate in the combination group was significantly higher than those in the chemotherapy group and the Chinese medicine group(all P<0.05).The expression levels of VEGF,Cyclin D1,Bcl-2 and Bcl-2/Bax ratio in the tumor tissues of the Chinese medicine group,chemotherapy group,and combination group were significantly lower than those of the model group(all P<0.05),and the expression level of Bax was significant higher than that of the model group(P<0.05);the expression levels of VEGF,Cyclin D1,Bcl-2,Bcl-2/Bax ratio in the tumor tissues of the combination group were significantly lower than those in the Chinese medicine group and the chemotherapy group(both P<0.05),the expression level of Bax was significantly highe
