摘 要:为揭示单核细胞增生李斯特菌(LM)lmo0732基因的分子特征及生物学功能,对LM lmo0732基因进行扩增、克隆和测序,利用在线软件对其进行分子特征及遗传进化分析。结果表明,lmo0732基因全长为1 917 bp,编码638个氨基酸;氨基酸序列分析发现,该蛋白为稳定的亲水性蛋白,含有1个跨膜区域,存在信号肽结构;蛋白结构域预测发现该蛋白含有Big 3、MucBp和LRR保守结构域;二、三级结构分析发现,该蛋白为无规则卷曲连接多段延伸链及α-螺旋所形成的三维立体结构。遗传进化分析显示,不同地域分离株可分为两个基因群,表现出一定的遗传多样性。In order to reveal the molecular characteristics and biological function of lmo0732, lmo0732 gene was amplified and cloned, and the sequence was analyzed by bioinformatics software and conducted to construct phylogenetic tree in this study. The results showed that the lmo0732 gene was successfully amplified with the length of 1917 bp and encoded 638 aa. Amino acid sequence analysis found that the Lmo0732 protein was a stable protein with hydrophilicity;it had a signal peptide and contained transmembrane domain. Protein domain prediction showed that the Lmo0732 protein included Big 3, MucBp and LRR domains;analysis on secondery and tertiary structure found that Lmo0732 protein was a three-dimensional structure formed by random coils connecting multiple extended strands and alpha helices. Phylogenetic analysis showed that isolates from different geographic regions could be divided into two gene groups and presented genetic diversity.
