摘 要:Gretchen hagen 3(GH3)属于植物生长素早期响应基因,通过调控生长素的稳态对植物的生长发育起着重要作用。然而,关于茶树GH3基因家族的研究尚缺乏系统报道。本文通过生物信息学的方法,在茶树全基因组数据中鉴定获得20个茶树GH3家族基因(CsGH3),并对其蛋白的理化性质、基因结构、保守基序等进行分析。理化性质分析结果表明,CsGH3基因家族编码的氨基酸分子量为27.5~87.1 kD,理论等电点为4.80~8.49,蛋白质不稳定性指数为32.40~49.58;基因结构分析结果表明,除CsGH3.16没有内含子外,其他CsGH3s都具有2~6个内含子;保守结构域分析结果显示,CsGH3家族主要含有10个Motif,编码的蛋白序列均含有Motif 1、Motif 2、Motif 4和Motif 6保守基序;系统进化树分析结果显示,茶树GH3基因家族可以分为两个亚家族,每个亚族所含基因数分别为11个和9个;启动子元件分析结果显示,GH3基因的启动子元件类型主要有激素响应元件、光响应元件、胁迫响应元件以及生长发育相关元件四大类;对茶树GH3基因在8个组织中的表达分析结果显示,20个基因在不同组织中的表达量呈现较大差异,表明这些基因可能在茶树不同的发育阶段起到重要作用。本文为进一步解析单个CsGH3基因的生物学功能提供参考。Gretchen hagen 3(GH3)is an early response gene of plant auxin,which plays an important role in plant growth and development by regulating auxin homeostasis.However,there is a lack of systematic report on the GH3 gene family in tea plant(Camellia sinensis).In this study,20 CsGH3 family genes were identified from the whole genome data of tea plant by bioinformatics method,and their protein physicochemical properties,gene structure and conserved motif were analyzed.The physicochemical properties analysis showed that the molecular weight of amino acids encoded by the CsGH3 gene family was 27.5~87.1 kD,the theoretical isoelectric point(PI)was 4.80~8.49,and the protein instability index was 32.40~49.58.Gene structure analysis showed that all CsGH3s had 2~6 introns except CsGH3.16,which had no introns.The results of the conservative domain analysis showed that the CsGH3 family mainly contained 10 motifs,and the encoded protein sequences contained Motif 1,Motif 2,Motif 4 and Motif 6.Phylogenetic plant analysis showed that the CsGH3 could be divided into two subfamilies,with the number of genes contained in each subfamily being 11 and 9,respectively.According to the results of promoter element analysis,there were four main types of promoter elements in CsGH3 gene:hormone response element,light response element,stress response element and growth and development related element.The results of the analysis of CsGH3 gene expression in 8 tissues of tea plant showed that the expression amount of 20 genes in different tissues showed great differences,indicating that these genes may play an important role in different development stages of tea plant.This study provides a reference for further analysis of the biological function of single CsGH3 gene.
