摘 要:采用TCGA公开的子宫内膜癌数据,分析微小染色体维持蛋白7基因(MCM7)在患者中的表达分布情况;采用交叉验证方法,将数据分为训练组和验证组,分析MCM7基因表达水平与放疗敏感性的关系。结果表明:MCM7基因表达在不同组织学和病理学分级上存在显著差异;MCM7基因高表达与患者放疗敏感性相关。在训练数据和验证数据中,MCM7基因高表达患者,在放疗和非放疗组生存差异有统计学意义,放疗患者的生存率显著提高,多因素校正后放疗的HR分别为0.337 (0.134~0.847)和0.312 (0.106~0.918),相应P值分别为0.021和0.034。合并训练组和验证组数据后,分析结果与上述一致。在全部放疗患者中,高表达患者生存率亦显著高于低表达患者,P值为0.024。不同组织学和病理学分级的亚组分析也与上述研究结果一致。这一研究提示MCM7基因高表达与放疗敏感性相关,MCM7基因可能是子宫内膜癌患者放疗敏感性潜在有效的生物标记物。We used the uterine corpus endometrial carcinoma data from TCGA to analyze MCM7 gene expression distribution in patients. The cross-validation procedure was used to study the association between the expression level of MCM7 and radio sensitivity, by randomly splitting the data to two parts, one as training data, and another one as validation data. The results showed that the expression level of MCM7 gene had significant differences in different histological stages and pathological grades. Furthermore, patients with high expression level of MCM7 gene were significantly associated with radio sensitivity. In training data and validation data, for patients with high expression of MCM7, significant better survival was observed for patients who received radiotherapy, compared with nonradio therapy patients. The adjusted HR was 0.337(0.134-0.847) and 0.312(0.106-0.918), with the P values 0.021 and 0.034 in training data and validation data, respectively. For the merged data from the training and validation dataset, the analysis results were consistent with the above. For all patients who received radiotherapy, the survival rate was significantly higher for patients with high expression level of MCM7 gene, compared patients with low gene level. The log-rank test P value was 0.024. The stratified analysis on histological stage and pathological grade showed the same conclusion. These results suggested that high expression level of MCM7 gene was strongly associated with radio sensitivity. The gene MCM7 might be a potential biomarker for radiotherapy prediction and treatment of uterine corpus endometrial carcinoma.
