摘 要:响应面法优化石豆兰总黄酮提取工艺,并考察醇提物抗氧化活性。单因素试验研究乙醇体积分数、液料比、温度和时间对总黄酮提取的影响,据单因素结果采用Box-Behnken法设计和响应面分析获取提工艺的数学模型和最优工艺;通过5个指标评价醇提物抗氧化活性。结果表明:最佳提取工艺为乙醇体积分数70%,液料比60∶1(mL/g),温度60℃,时间25 min,此条件下总黄酮实际浸提量为24.85 mg/g,与理论值24.95 mg/g基本符合;醇提物的总还原力是Vc还原力的39.68%,清除DPPH自由基、羟基自由基和超氧阴离子的能力分别是Vc的53.40%、21.70%、50.40%,对Fe2+的螯合力是EDTA的1.2%。表明石豆兰醇提物具较好的抗氧化能力,可以进行功能性食品或药品的开发。In this experiment,optimization of extraction of total flavonoids from Bublophyllum kwangtungense and its antioxidant activity were studied.Firstly,ethanol concentration,liquid-to-solid,extraction temperature and time were selected as factors to conduct single factor investigations for the purpose of establishing the appropriate levels.Then,a predictive mathematical model was developed using Box-Behnken experimental design for the optimum process by response suface analysis.And the antioxidant activity of ethanol extraction was evaluated by five indexes.The results showed that the optimum process were obtained as follows:Ethanol concentration,70%;Liquid-to-solid,60∶1(mL/g);Extraction temperature,60℃;And extraction time,25 min.Under the optimum process,the actual yield of total flavonoids was 24.85 mg/g,which was close to the predicted value of 24.95 mg/g.The total reducing power of ethanol extraction was 39.68%of Vc.The scavenging capabilities for DPPH,hydroxyl radicals and superoxide anions were 53.40%,21.70%,50.40%of Vc respectively.And the chelating rate to Fe2+was 1.20%of EDTA.The-refore,the ethanol extraction from B.kwangtungense had strong antioxidant capacity and could be develop functional food or medicine.
