摘  要:为研究敲低分泌型磷蛋白1(SPP1)基因对人口腔鳞状细胞癌(OSCC)细胞株HSC-3细胞凋亡的影响,本研究从基因表达综合(GEO)数据库下载表达谱芯片数据GSE31056,用Bioconductor中的Limma包筛选差异表达基因。利用靶向设计并构建SPP1重组质粒载体,转染HSC-3细胞,Real-time PCR和Western blotting分别检测SPP1基因和蛋白表达情况,细胞流式术检测凋亡率,Western blotting检测PI3K/AKT信号通路蛋白表达水平。筛选出的差异表达基因中SPP1在OSCC中高表达。敲低SPP1后其基因和蛋白表达水平明显下降,细胞凋亡率明显增加,PI3K/AKT信号通路蛋白表达水平下降。敲低SPP1通过PI3K/AKT信号通路促进OSCC HSC-3细胞凋亡。To investigate the effect of knockdown of secreted phosphoprotein 1(SPP1)gene on the apoptosis of human Oral squamous cell carcinoma(OSCC)HSC-3 cell.GSE31056 was downloaded from Gene Expression Omnibus(GEO)which is the m RNA expression profiles dataset and the differentially expressed genes(DEGs)were screened by limma package which is belong to Bioconductor in RStudio.Targeting the design and construction of SPP1 recombinant plasmid vector and transfecting it into HSC-3 cells.The expression of SPP1 gene and protein were detected by real-time PCR and Western blot.The apoptosis rate was detected by flow cytometry.The protein expression of PI3 K/AKT signaling pathway was detected by Western blotting.A large number of differentially expressed genes were screened,of which SPP1 was highly expressed in oral squamous cell carcinoma(OSCC).After knocking down SPP1,the expression levels of the gene and protein decreased significantly,the apoptosis rate increased significantly,and the expression level of PI3 K/AKT signaling pathway protein decreased.Knockdown of SPP1 gene promotes apoptosis of oral squamous cell carcinoma cell line HSC-3 through the PI3 K/AKT signaling pathway.
