摘 要:为了探讨硫化氢对癫痫大鼠脑电图改变和炎症因子的影响,本研究通过腹腔注射戊四氮诱导大鼠癫痫持续状态模型,然后腹腔注射硫氢化钠溶液对癫痫大鼠进行预处理。研究发现,硫氢化钠处理可显著升高大鼠的血浆(102.19102.15μmol/L)和海马组织中的硫化氢浓度,但未超过动物体内硫化氢的生理浓度。与模型组大鼠相比,硫氢化钠处理可显著延长大鼠癫痫发作至Ⅳ级时的潜伏期(31.86 min vs 16.24 min),并显著降低癫痫持续时间(65.85 s vs 118.93 s)。在模型组大鼠脑电图中可观察到明显的尖棘波或棘慢波,振幅明显升高(536.255μV)。硫氢化钠处理则可显著减少尖棘波或棘慢波,并降低振幅(361.58μV)。尼式染色法显示,模型组的神经元明显变形,数量减少,染色较浅,尼氏小体数量明显减少。而硫氢化钠处理组的神经元数量与模型组相比有所增加,且完整性明显改善,尼氏小体数量增多。RT-qPCR和Western blotting结果显示,硫氢化钠处理可显著降低TNF-α、IL-1β、IL-6和NF-κB/p65的mRNA和蛋白相对表达量。本研究证实,在硫化氢的生理浓度范围内,增加硫化氢浓度可通过抑制NF-κB信号通路的激活来降低炎症反应,从而发挥抗癫痫的作用。In order to investigate the effects of hydrogen sulfide on EEG changes and inflammatory factors in rats with epilepsy,this study used a model of epilepticus status induced by intraperitoneal injection of pentylenetetrazol.The epileptic rats were then pretreated by intraperitoneal injection of sodium hydrosulfide solution.The study found that sodium hydrosulfide treatment significantly increased the concentration of hydrogen sulfide in plasma(102.19~102.15μmol/L)and hippocampus of rats,but did not exceed the physiological concentration of hydrogen sulfide in animals.Compared with the model group,sodium hydrosulfide treatment significantly prolonged the incubation period of seizures to gradeⅣ(31.86 min vs 16.24 min)and significantly reduced the duration of epilepsy(65.85 s vs 118.93 s).Significant spike or spine waves were observed in the EEG of the model group,and the amplitude was significantly increased(536.255μV).Sodium hydrosulfide treatment significantly reduced sharp spikes or spine slow waves and reduced amplitude(361.58μV).Nicholas staining showed that the neurons in the model group were significantly deformed,the number was reduced,the staining was shallow,and the number of Nissl bodies was significantly reduced.The number of neurons in the sodium hydrosulfide treatment group was increased compared with the model group,and the integrity was significantly improved,and the number of Nissl bodies was increased.RT-qPCR and Western blotting showed that sodium hydrosulfide treatment significantly reduced the mRNA and protein expression levels of TNF-α,IL-1β,IL-6 and NF-κB/p65.This study demonstrates that increasing the concentration of hydrogen sulfide in the physiological concentration range of hydrogen sulfide can reduce the inflammatory response by inhibiting the activation of the NF-κB signaling pathway,thereby exerting an antiepileptic effect.
