摘  要:为了揭示有氧运动对脑缺血大鼠神经功能和认知功能的改善作用机制,本研究建立了大鼠中脑动脉闭塞手术(MCAO)诱导缺血性中风大鼠模型。将大鼠在跑步机上以10 m/min的速度运动30 min,每天运动1次,共运动4周。用2,3,5-三苯四唑氯化物(TTC)染色评估MCAO手术是否成功建立。通过神经功能缺损评分来考察大鼠的神经功能、Morris水迷宫实验考察大鼠的认知功能、免疫荧光染色检测大鼠脑缺血半影区中的神经发生和血管生成。此外,通过蛋白质印迹法测定缺血半影区中的TLR4、p-NF-κBp65、NF-κBp65、NLRP3和IL-1β的蛋白水平。结果表明,有氧运动不仅降低了大鼠的脑梗死面积和神经功能缺损评分(p<0.05),还降低了大鼠的逃避潜伏期,提高了穿越平台次数(p<0.01)及增加了缺血性中风后新生神经元的数量和脑微血管密度(p<0.05)。此外,有氧运动下调了大鼠脑缺血半影区中TLR4、p-NF-κBp65、NF-κBp65、NLRP3和IL-1β的蛋白水平(p<0.05)。研究发现,有氧运动可抑制TLR4/NF-κB/NLRP3信号传导途径来促进大鼠脑缺血半影区的神经发生和血管生成,从而提高脑缺血大鼠的神经功能和认知功能。In order to reveal the mechanism of aerobic exercise to improve the neurological and cognitive functions of cerebral ischemic rats.This study included a rat model of ischemic stroke induced by middle cerebral artery occlusion surgery(MCAO).The rats were exercised at a speed of 10 m/min for 30 min on a treadmill.Exercise once a day for 4 weeks.The 2,3,5-triphenyltetrazolium chloride(TTC)staining was used to evaluate the successful establishment of MCAO surgery.The neurological function of rats was examined by the neurological deficit score.The cognitive function of rats was investigated through the Morris water maze experiment.Immunofluorescence staining was used to detect neurogenesis and angiogenesis in the ischemic penumbra of rats.In addition,the protein levels of TLR4,p-NF-κBp65,NF-κBp65,NLRP3 and IL-1βin the ischemic penumbra were determined by Western blotting.The results showed that aerobic exercise not only reduced the cerebral infarction area and neurological deficit score of rats(p<0.05),but also reduced the escape latency of rats and increased the number of crossing platforms(p<0.01),and increased the number of new neurons and the density of cerebral micro vessels after ischemic stroke(p<0.05).In addition,aerobic exercise reduced the protein levels of TLR4,p-NF-κBp65,NF-κBp65,NLRP3 and IL-1βin the penumbra of cerebral ischemia in rats(p<0.05).This study showed that aerobic exercise can inhibit the TLR4/NF-κB/NLRP3 signaling pathway to promote neurogenesis and angiogenesis in the ischemic penumbra of rats,thereby improving the neurological and cognitive functions of rats with cerebral ischemia.
