摘 要:目的探讨EGCG-CoPt NPs-Apt纳米粒子对三阴乳腺癌细胞MDA-MB-231增殖及迁移的影响。方法利用合成的EGCG-CoPt纳米粒子,偶联EGCG及核酸适配体AS1411来构建EGCG-CoPt NPs-AS1411纳米粒子,分别采用亚甲蓝法及Transwell法观察其对三阴乳腺癌细胞MDA-MB-231增殖及迁移的影响,最后采用Western blot检测MDA-MB-231细胞中肿瘤转移相关因子MMP-2、MMP-9、EGFR和VASP的蛋白表达水平。结果成功构建复合型纳米粒子EGCGCoPt NPs-AS1411,与CoPt NPs组、对照组及EGCG-CoPt NPs相比,EGCG-CoPt NPs-AS1411能显著抑制三阴乳腺癌细胞MDA-MB-231的增殖、迁移及MMP-9、EGFR和VASP蛋白表达,差异有统计学意义(P<0.05)。结论EGCG-CoPt NPs-Apt纳米粒子能充分利用纳米粒子和核酸适配体的双重靶向作用,进一步发挥EGCG及核酸适配体AS1411抑制乳腺癌细胞增殖、迁移的能力,抑制三阴乳腺癌细胞MDA-MB-231的增殖与转移。Objective To investigate the effect of EGCG-CoPt NPs-Apt nanoparticles on the proliferation and migration of triple negative breast cancer cell MDA-MB-231.Methods EGCG-CoPt NPs-AS1411 nanoparticles were synthesized by coupling EGCG with aptamer AS1411.The effects of EGCG-CoPt NPs-AS1411 on proliferation and migration of triple negative breast cancer cells MDA-MB-231 were observed by methylene blue method and transwell method respectively.Finally,western blot was used to detect the protein expression of MMP-2,MMP-9,EGFR and VASP in MDA-MB-231 cells.Results The composite nanoparticles EGCG-CoPt NPs-AS1411 was successfully constructed.Compared with CoPt NPs group,control group and EGCG CoPt NPs group,the proliferation and migration of MDA-MB-231 cells as well as the expression of MMP-9,EGFR and VASP proteins were significantly inhibited by EGCG-CoPt NPs-AS1411(P<0.05).Conclusion EGCG-CoPt NPs-AS1411 nanoparticles can make full use of the dual targeting effect of nanoparticles and aptamers,further exert the ability of EGCG and aptamer As1411 to inhibit the proliferation and migration of breast cancer cells,and inhibit the proliferation and metastasis of triple negative breast cancer cells MDA-MB-231.
