摘  要:目的探讨一种雌性犬尿道壁内血管窦压力的测定方法。方法健康成年雌性犬10条,购自华中科技大学同济医学院附属同济医院实验动物中心。麻醉下显露膀胱及尿道外口并在膀胱、尿道高压段腔内和尿道壁血管窦内放置测压管用于测定膀胱内压(IBP)、尿道高压段腔内定点压(IUP)和尿道壁内血管窦压(VSP)。每条犬分别在静息和应力状态同步测定IBP、IUP及VSP。完成上述测压后,经VSP测压通道灌注肝素墨汁混合液,过量麻醉处死犬取全尿道和膀胱标本固定后行冰冻切片观察尿道壁内血管窦墨汁充盈着色,以验证VSP所反映的血管窦结构。采用SPSS 19.0统计软件分析,实验数据使用计量资料的统计描述,以均值±标准差(Mean±SD)表示。结果本方法能够准确记录静息状态下稳定的VSP值和应力状态下灵敏的VSP变化,并实现IBP、IUP、VSP的同步测定。本组10条成年雌性犬静息状态下IBP、IUP、VSP分别为(9.10±2.81)、(21.02±3.83)、(12.27±3.51)cmH2O(1 cmH2O=0.098 kPa),IUP/VSP为1.86±0.64;应力状态下,IBP、IUP、VSP分别为(43.05±6.41)、(52.01±7.47)、(17.80±4.50)cmH2O,IUP/VSP为3.08±0.81。墨汁灌注冰冻切片可见各节段尿道壁内血管窦被墨汁充盈染色,从形态学角度证实本研究所测的VSP为尿道壁内血管窦内压力。结论本研究建立了一种准确测定雌性犬尿道壁内血管窦压力的方法,为进一步研究尿道压力的形成机制奠定了方法学基础。Objective To describe a method for measuring the vascular sinusoids pressure(VSP)of urethral wall in female canines.Methods Totally 10 adult healthy female canines used for this study.All animals were purchased from experimental animal center of Tongji hospital.The urinary bladder,urethral meatus were exposed under intravenous pentobarbital anesthesia and cannulated for measuring the intra bladder pressure(IBP),intraurethral pressure(IUP)at the maximum urethral pressure zone and vascular sinusoids pressure(VSP)at the distal urethral wall.Then the IBP,IUP and VSP were recorded synchronously at rest and stress status.Subsequently,heparin sodium ink mixture solution was infused into the vascular sinusoids via the VSP tube,then the whole urethra and bladder were removed en block for frozen section,which providing morphological evidence for the accuracy of the VSP measuring system.The data analysis was conducted by SPSS 19.0,the quantitative data were expressed as the means±tandard deviation(Mean±SD).Results The VSP measuring system established in the present study can obtained a reliable static baseline pressure at rest status and sensitive synchronous changes with IBP and IUP at stress status.At rest status,VSP,IUP,IBP were(12.27±3.51),(21.02±3.83),(9.10±2.81)cmH2O(1 cmH2O=0.098 kPa)respectively and IUP/VSP was 1.86±0.64.At stress status,VSP,IUP、IBP were(17.80±4.50),(52.01±7.47),(43.05±6.41)cmH2O respectively and IUP/VSP was 3.08±0.81.The urethral vascular sinusoids stained with ink were observed in each urethral zone,which provided direct morphological evidence that the VSP obtained in this present study was the intra-sinusoidal pressure.Conclusion A method of direct measuring VSP of the urethral wall in female canine was successfully established for the first time,that may play important role in further investigation of the vascular components in urethral closure mechanisms.
