摘  要:建立了大米中水溶性糖的测定方法。大米中的水溶性糖用热水提取,乙酸锌、亚铁氰化钾溶液沉淀,经盐酸水解后用3,5-二硝基水杨酸显色,分光光度计比色测定。结果表明,在3,5二硝基水杨试剂用量1.0 mL,沸水浴加热6 min,540 nm波长条件下进行测定,标准曲线线性关系良好,回归方程Y=0.9682 X+0.0144,相关系数R 2=0.9996,在0.1%、0.5%和2.0%三个标准添加浓度下,水溶性糖的平均加标回收率为91.9%~98.3%,相对标准偏差为2.68%~2.79%,方法检出限为0.003%。该方法准确度和灵敏度高,重现性好,操作简单、快速,有应用价值。10份不同品种大米水溶性糖含量测定结果为0.12%~1.95%。A method for determination of water-soluble sugar in rice was established.The water-soluble sugar in rice was extracted by hot water,and purified with zinc acetate dehydrate and potassium ferrocyanide.The water-soluble sugar in rice was coloration with 3,5-dinitrosalicylic acid,and analysis with spectrophotometer colorimeter.The result indicated that,under the conditions of 1 mL 3,5-dinitrosalicylic acid usage,6 minutes boiling water heating,540 nm wavelength determination,the standard curves had good linearity,linear regression equation was Y=0.9682X+0.0144,correlation coefficient R 2=0.9996.Under adding concentration of 0.1%,0.5%,2.0%,the average recoveries were 91.9%~98.3%,the relative standard deviation was 2.68%~2.79%.The detection limit of this method was 0.003%.The established method has the advantages of simply,rapidity,high accuracy and sensitivity,and was suitable for the determination of water-soluble sugar in rice.The determination results of water-soluble sugar in 10 rice varieties were 0.12%~1.95%.
