LncRNA Neat1在肝细胞癌组织和细胞株中的表达及临床意义

摘　　要：目的探讨长链非编码RNA(LncRNA)Neat1在肝细胞癌组织及细胞株中的表达及临床意义。方法选取2016年1月至2017年12月在浙江省台州医院肝胆外科接受手术的16例肝细胞癌患者为研究对象,收集所有患者癌组织及癌旁正常肝组织(距肿瘤2 cm以上)标本。同时培养正常人肝细胞(LO2)、人高转移肝癌细胞(MHCC97-H)、人低转移肝癌细胞(MHCC97-L)。采用RT-qPCR法检测肝细胞癌组织、癌旁正常组织及MHCC97-H、MHCC97-L、LO2细胞株LncRNA Neat1相对表达量,比较癌组织与癌旁正常组织、肝癌细胞株与正常人肝细胞的表达差异,分析肝细胞癌患者临床特征与癌组织中LncRNA Neat1相对表达量的关系,利用ROC曲线分析LncRNA Neat1相对表达量诊断肝细胞癌的效能。结果肝细胞癌组织LncRNA Neat1相对表达量明显低于癌旁正常组织(P<0.05),且与肿瘤分化程度有关(P<0.05),与患者年龄、性别、肿瘤部位以及血清甲胎蛋白、ALT、AST、TBil水平均无关(P>0.05)。LncRNA Neat1相对表达量诊断肝细胞癌的AUC、约登指数、灵敏度、特异度分别为0.793、0.500、0.875、0.625。2种肝癌细胞株LncRNA Neat1相对表达量均明显低于正常人肝细胞(均P<0.05),而2种肝癌细胞株的表达量比较差异无统计学意义(P>0.05)。结论肝细胞癌组织及肝癌细胞株中LncRNA Neat1表达明显降低,肝细胞癌组织LncRNA Neat1表达与肿瘤分化程度密切相关。LncRNA Neat1有望成为诊断肝细胞癌的新型肿瘤标志物。Objective To explore the expression and clinical significance of LncRNA Neat1 in HCC.Methods A total of 16 patients with hepatocellular carcinoma who underwent surgery in the Department of Hepatobiliary Surgery,Taizhou Hospital,Zhejiang Province from January 2016 to December 2017 were selected as research objects,and samples of cancer tissues and adjacent normal liver tissues(more than 2 cm away from the tumor)were collected from all patients.Meanwhile,normal hepatocytes(LO2),human hepatocellular carcinoma cells with high metastasis(MHCC97-H),and human hepatocellular carcinoma cells with low metastasis(MHCC97-L)were cultured.The relative expression levels of LncRNA Neat1 in hepatocellular carcinoma tissues,adjacent normal tissues and MHCC97-H,MHCC97-L and LO2 cell lines were detected by RT-qPCR,to compared the expression differences between carcinoma tissues and paracancer normal tissues,and between hepatocellular carcinoma cell lines and normal human hepatocytes,to analyzed the relationship between the clinical characteristics of HCC patients and the relative expression of LncRNA Neat1 in HCC tissues,ROC curve was used to analyze the efficacy of LncRNA Neat1 in the diagnosis of HCC.Results The relative expression level of LncRNA Neat1 in hepatocellular carcinoma tissues was significantly lower than that in adjacent normal tissues(P<0.05),and was related to the degree of tumor differentiation(P<0.05).It was not related to the age,gender,tumor site,serum levels of a-fetoprotein,ALT,AST,and TBil of patients(P>0.05).The AUC,Yoden index,sensitivity and specificity of the relative expression of LncRNA Neat1 in the diagnosis of HCC were 0.793,0.500,0.875 and 0.625,respectively.The relative expression levels of LncRNA Neat1 in the two hepatocellular carcinoma cell lines were significantly lower than that in normal human hepatocytes(all P<0.05),while the difference in the expression levels of the two hepatocellular carcinoma cell lines was not statistically significant(P>0.05).Conclusion The expression of LncRNA Neat1 in
