摘 要:目的:筛选并鉴定高脂血症性胰腺炎(HLP)患者治疗前后血清差异表达的蛋白质。方法:采用同位素标记相对和绝对定量(iTRAQ)技术结合液相串联质谱(2DLC-MS/MS)检测30例HLP患者治疗后与治疗前血清差异表达的蛋白质,进行3组重复试验,并筛选差异蛋白进行分析。结果:共鉴定出蛋白质367种,与HLP治疗前组比较,H P治疗后组共获得显著差异蛋白91个(差异倍数>1.2或<0.83,且P<0.05),其中上调78个,下调13个。结论:血清中鉴定获得的差异蛋白MASP2、CLUS可能是提高HLP治疗效果的潜在蛋白,SAA1可能成为预后随访的指标,值得深入研究。Objective To screen and identify the differential expression of serum proteins in patients with hyperlipidemic pancreatitis(HLP)before and after treatment.Methods Isobaric tags for relative and absolute quantification(iTRAQ)and two-dimensional liquid chromatography-tandem mass spectrometry(2DLC-MS/MS)were used to detect the differentially expressed proteins in serum of 30 patients with HLP after treatment and before treatment.Three groups of repeated tests were carried out and the differential proteins were screened for analysis.Results A total of 367 proteins were identified.Compared with the HLP pre-treatment group,91 proteins were significantly different after HLP treatment(fold>1.2 or fold<0.83,and P<0.05),including 78 up-regulated proteins and 13 down-regulated proteins.Conclusion The differential protein MASP2 and CLUS identified in serum may be the potential protein to improve the therapeutic effect of HLP.SAA1 may be an indicator of prognosis and follow-up,which is worthy of further study.
