摘  要:探讨一套能快速、准确定位脑区的操作方法,以解决传统实验动物因体型大小、性别差异以及新兴实验动物因缺乏相应图谱和立体定位坐标信息而难以精确定位脑区的问题。其主要方法是以长爪沙鼠(Meriones unguiculatus)为研究对象,先行切片制作出一套与待进针体型相近的同性长爪沙鼠的尼氏染色脑图谱,比对小鼠脑图谱以初步了解目的脑区周围结构及相对位置。再利用桌面数显型脑立体定位仪固定其头部,并以前囟为原点垂直进针以在脑组织留下进针痕迹,快速断头后制备脑组织冠状冰冻切片。以尼氏染色脑图谱为基础,测量与计算目标脑区与前囟的相对位置,初步推算三维空间坐标。进针此坐标并再次制备切片并计算进针点与目标脑区的偏差值,即可得到该脑区的精确坐标,重复上述步骤验证该坐标数据的精确度。本实验通过上述方法快速进针左右两侧若干脑区并获得其坐标数据,有左右两侧侧脑室(LV),室旁核(PVH),中央杏仁核(CeA),内侧杏仁核(MeA),终纹床核(BST)等研究热点脑区,均有较高的成功率,因此该方法适用于快速准确定位脑区。We tested methods for quickly and accurately locating brain regions to solve the problems of traditional experimental animals for which it is difficult to accurately locate brain regions due to size and gender differences.We also aimed to identify target brain regions of species of animals whose importance is now growing in experimental research.In this experiment,Meriones unguiculatus was used as the research species.Firstly,a set of gerbil Nissl-stained brain atlases was made using gerbils of similar size and of the same gender to understand the location of brain regions and to locate the brain regions accurately and handily.We used a dual arm digital display brain stereotaxic instrument to fix the gerbil head.The anterior fontanel was used as the origin,and was vertically inserted by the needle to leave a needle mark on the brain tissue.Frozen corona-sections of brain were performed after a rapid decollation.Based on the Nissl-stained brain map atlases,the three-dimensional coordinates of the brain region were preliminarily estimated by measuring and calculating the relative positions of the target brain region and the anterior fontanel.According to the estimated coordinates,another brain was inserted by needle and made into frozen sections.The deviation value between the internal fixation point and the target brain region was calculated,then the precise coordinates of the brain region were obtained.These steps were repeated for verification based on the exact coordinates.Using this method,the three-dimensional coordinates of the left and right brain regions of Mongolian gerbils were accurately located,including the lateral ventricle(LV),the paraventricular nucleus(PVH),the central amygdala nucleus(CeA),the medial amygdala nucleus(MeA),and the bed nucleus of the stria terminalis(BST).This method proved suitable for the quick and accurate localization of brain regions.
