摘 要:通过分析空白组、模型组、盐酸硫必利组的刻板行为和其脑匀浆中含多巴胺量的关系,建立评价抽动-秽语综合征模型小鼠刻板行为程度的数学模型,并对该数学模型进行验证探讨.取24只清洁级ICR小鼠随机分为空白组、模型组和盐酸硫必利组,每组8只.通过持续15 d腹腔注射3,3’-亚氨基二丙腈建立抽动症模型,盐酸硫必利组小鼠每日按10 g/kg灌胃盐酸硫必利片混悬液,测第21天刻板行为和脑匀浆中含多巴胺量.通过三组小鼠的刻板行为发生频率与其脑匀浆中含多巴胺量进行多元线性回归分析,并建立数学模型.小鼠刻板行为测试结果显示与空白组相比,模型组的口爪运动显著增加(P<0.05),点头、自咬、摆头、旋转非常显著增加(P<0.01);与模型组相比,硫必利组点头、口爪运动显著降低(P<0.05),自咬、摆头非常显著降低(P<0.01).数据分析求得该数学模型为y=368.256-10.122x1-13.696x2-96.742x3+20.907x4+56.738x5-81.969x6+1.431×10-8ex1+0.371ex2-3.621ex3+0.037ex4-0.388ex5-4.028ex6-0.192x12+27.640x32-7.602x42+38.861x62+47.548x72.该数学模型评价刻板行为效果较好,能将抽动-秽语综合征模型鼠所表现出的多种刻板动作相联系,科学、直观、完整地评价和比较不同抽动症模型鼠刻板行为的严重程度.Analyzing the relationship between stereotypical behavior and dopamine content in brain homogenate of blank group,model group and tiapride hydrochloride group to establish a mathematical model to evaluate the degree of stereotyped behavior in mice with Tourette syndrome.Discussed and verified mathematical model.Took twenty four ICR mice and randomly divided into blank group,model group and thiopride hydrochloride group with 8 mice in each group.Kept the intraperitoneal injection of 3,3’-iminodipropiononitrile to model of TiC for 15 days.The rats in the group of tiapride hydrochloride were administrated with the suspension of tiapride hydrochloride tablets at the dose of 10 g/kg per day.And measure the stereotypical behavior and dopamine content in brain homogenate on the 21 st day.Established a mathematical model by multiple linear regression analysis to the frequency of stereotyped behavior and dopamine content in brain homogenate of three groups of mice.The results of the stereotyped behavior test showed that compared with the blank group,the movement of jaws in the model group increased significantly(P <0.05),and that of nodding,self biting,head swinging and rotation increased significantly(P <0.01);compared with the model group,the movement of nodding and jaws in the thiopride group decreased significantly(P <0.05),and that of self biting and head swinging decreased significantly(P <0.01).It was analyzed by data that the mathematical model was y=368.256-10.122 x1-13.696 x2-96.742 x3+20.907 x4+56.738 x5-81.969 x6+1.431×10-8 ex1+0.371 ex2-3.6216x3+0.037 ex4-0.388 ex5-4.028 ex6-0.192 x1227.640 x32-7.602 x42+38.861 x62+47.548 x72.The mathematical model has a good effect on the evaluation of stereotyped behavior.It can be used to evaluate and compare the severity of stereotyped behavior scientifically,intuitively and completely.
