原花青素与维生素B1对酒精致小鼠大脑β淀粉样蛋白过量产生的影响

摘  要:目的探讨酒精摄入对小鼠大脑β淀粉样蛋白1-42(Aβ1-42)、β淀粉样蛋白1-40(Aβ1-40)、β淀粉样前体蛋白(β-APP)的影响以及原花青素(PC)与维生素B1(VB1)的营养干预作用。方法 C57BL/6小鼠随机分为对照组、0.5%、1%、2%、3%、4%酒精组,PC+4%酒精组、VB1+4%酒精组、PC+VB1+4%酒精组,酒精液体饲料配对喂养10周后,检测脑Aβ1-42、Aβ1-40、β-APP的含量及其在大脑皮层的表达情况。结果与对照组相比,3%、4%酒精组小鼠大脑Aβ1-42、Aβ1-40、β-APP的含量均明显升高,2%酒精组小鼠大脑Aβ1-40的含量也明显升高;与4%酒精组相比,PC+VB1+4%酒精组小鼠脑中Aβ1-42、Aβ1-40、β-APP的水平明显降低,PC+4%酒精组Aβ1-42与β-APP的含量明显降低;PC与VB1对脑Aβ1-40含量的影响有交互作用。另外,摄入2%、3%、4%酒精的小鼠大脑皮层Aβ1-42、Aβ1-40的表达明显增加,PC与VB1合用能明显减少Aβ1-42、Aβ1-40的表达。结论 PC与VB1合用通过抑制Aβ在小鼠脑中的过量生成发挥对酒精性脑损伤的保护作用。Aim To investigate the effects of ethanol(EtOH)consumption onβ-amyloid peptide 1-42(Aβ1-42),β-amyloid peptide 1-40(Aβ1-40),andβ-amyloid precursor protein(β-APP)levels in brain of mice,and to study the nutritional intervention of proanthocyanidins(PC)and vitamin B1(VB1)on alcoholic mice.Methods C57BL/6 mice were randomly divided into nine groups:control,0.5%EtOH,1%EtOH,2%EtOH,3%EtOH,4%EtOH,PC+4%EtOH,VB1+4%EtOH,and PC+VB1+4%EtOH.After paired-feeding with an EtOH liquid diet for ten weeks,the contents of brain Aβ1-42,Aβ1-40,andβ-APP levels were measured,and the expression levels of Aβ1-42 and Aβ1-40 in the cortex were observed.Results Compared with control group,the contents of Aβ1-42,Aβ1-40,andβ-APP in brain of 3%and 4%EtOH group mice significantly increased.The content of Aβ1-40 in brain of 2%EtOH group mice was significantly higher than that of control group.Compared with 4%EtOH group,the levels of Aβ1-42,Aβ1-40,andβ-APP in brain from PC+VB1+4%EtOH group mice significantly decreased,and the level of Aβ1-42 andβ-APP from PC+4%EtOH group′s brain also significantly decreased.The interaction effect of PC and VB1 on the reduction in Aβ1-40 level was significant.In addition,the expression levels of Aβ1-42 and Aβ1-40 in cerebral cortex of mice that consumed 2%,3%,and 4%EtOH markedly increased,and the combined use of PC and VB1 could significantly reduce the expressions.Conclusions The combination of PC and VB1 has a protective effect on alcohol-induced brain injury by inhibiting excessive production of Aβin mouse brain.
