摘  要:以副溶血性弧菌(Vibrio parahemolyticus)为指示菌,采用滤纸片法测定抗菌活性。通过乙酸乙酯萃取、硅胶吸附柱层析、薄层层析(TLC)及高效液相色谱(HPLC)等分离纯化技术,对来自海洋链霉菌HS-B34菌株发酵液中的抗菌活性物质进行分离纯化。研究结果发现,发酵液的乙酸乙酯粗提物中含有4种组分(A1~A4),其中A1和A2两组分有抗菌活性,而A3和A4无抗菌活性。进一步采用质谱及傅里叶红外光谱技术对活性较强含量较多的A1组分进行结构分析与鉴定,确认该组分为邻苯二甲酸二丁酯(DBP)。该组分A1对食品加工和海水养殖中多种常见的病原菌具有光谱抗菌活性,尤其对副溶血性弧菌具有显著的抑菌作用,显示出良好的生防应用潜力。Taking Vibrio parahemolyticus as the indicator bacteria,the antibacterial activity was determined by the filter paper method.Separation and purification techniques such as ethyl acetate extraction,silica gel adsorption column chromatography,thin layer chromatography(TLC)and high performance liquid chromatography(HPLC)were used to separate antibacterial active substances from the fermentation broth of Streptomyces marinus HS-B34 purification.The results indicated that the crude ethyl acetate extract of the fermentation broth contained 4 components(A 1~A 4),among them A 1 and A 2 had antibacterial activity,while A 3 and A 4 had no antibacterial activity.Furthermore,mass spectrometry and Fourier infrared spectroscopy were used to analyze and identify the more active A 1 component and it was confirmed that the component was dibutyl phthalate(DBP).The component A 1 had spectrum antibacterial activity against a variety of common pathogenic bacteria in food processing and marine aquaculture,especially the significant antibacterial effect on Vibrio parahaemolyticus,showing good application potential for biocontrol.
