摘　　要：目的探讨YAP基因对HPV感染宫颈癌细胞转化生长因子α(TGF-α)及表皮生长因子受体(EGFR)表达的调节作用。方法免疫组化及Western blot检测HPV-16(+)Ca Ski、HPV-18(+)C4-1人宫颈癌细胞,HPV(-)C33a人宫颈癌细胞中YES相关蛋白(YAP)、TGF-α及EGFR的表达。给予YAP抑制剂Vertepofin干预后,对Ca Ski及C4-1细胞及C33a增殖影响及对YES相关蛋白(YAP)、TGF-α、EGFR中mRNA及蛋白表达的影响。结果HPV-16(+)Ca Ski、HPV-18(+)C4-1人宫颈癌细胞中YAP、TGF-α、EGFR表达高于HPV(-)C33a细胞;给予Vertepofin干预后,细胞增殖速度降低,在YAP被抑制的同时,TGF-α、EGFR中mRNA及蛋白的表达也出现了抑制。结论HPV感染宫颈癌患者细胞中EGFR以及TGF-α表达Hippo/YAP信号通路可以起到正调控的作用。Objective To investigate the regulatory effect of YAP gene on the expression of TGF-αand EGFR in cervical cancer cells infected with HPV.Methods Immunohistochemistry and Western blot were used to detect the expression of YAP,TGF-αand EGFR in HPV-16(+)Ca Ski,HPV-18(+)C4-1 human cervical cancer cells and HPV(-)C33 a human cervical cancer cells.After intervention with the YAP inhibitor Vertepofin,it affected the proliferation of Ca Ski and C4-1 cells and C33 a,as well as the expression of mRNA and protein in YAP,TGF-α,and EGFR.Results The expressions of YAP,TGF-αand EGFR in HPV-16(+)Ca Ski and HPV-18(+)C4-1 human cervical cancer cells were higher than that of HPV(-)C33 a cells;after Vertepofin intervention,the cell proliferation rate decreased.At the same time that YAP was inhibited,the expression of mRNA and protein in TGF-αand EGFR also appeared to be inhibited.Conclusion The Hippo/YAP signaling pathway for EGFR and TGF-αexpression in HPV-infected cervical cancer cells can play a positive regulatory role.
