欧前胡素对脑缺血再灌注大鼠海马区损伤及ATP酶活力的影响

摘　　要：目的研究欧前胡素对脑缺血再灌注大鼠海马区损伤及ATP酶活力的影响。方法建立脑缺血再灌注大鼠模型,将大鼠随机分为6组:假手术组、模型组、欧前胡素25、50、100 mg/kg组和尼莫地平组进行后续实验,Y迷宫实验检测大鼠脑部学习记忆功能;HE染色检测大鼠脑组织病理损伤;TUNEL染色检测大鼠神经元细胞凋亡;试剂盒检测总超氧化物歧化酶(SOD)含量、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH)含量;试剂盒检测乳酸脱氢酶(LDH)、Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶活力;Western blot检测Survivin、Caspase-3、Caspase-9蛋白表达水平。结果与模型组相比较,假手术组及欧前胡素50、100 mg/kg组和尼莫地平组新异臂进入次数升高(P<0.05),新异臂潜伏期、起始臂进入次数、其他臂进入次数降低(P<0.05),病理损伤程度改善,凋亡阳性细胞数减少,Survivin蛋白水平升高(P<0.05),Caspase-3、Caspase-9蛋白水平降低(P<0.05),SOD、GSH含量升高(P<0.05),MDA含量降低(P<0.05),LDH活力降低(P<0.05),Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶活力升高(P<0.05)。结论欧前胡素对脑缺血再灌注模型大鼠具有保护作用。Objective To investigate the effects of imperatorin on hippocampal injury and atpase activity in rats after cerebral ischemia and reperfusion. Methods The rat model of cerebral ischemia-reperfusion was established,and the rats were randomly divided into 6 groups: healthy control group, model group, imperatorin 25,50,100 mg/kg group and nimodipine group for follow-up experiments. Y maze experiment detected the learning and memory function of rat brain. HE staining was used to detect pathological damage of brain tissue in rats. Apoptosis of rat neurons was detected by TUNEL staining. The contents of total superoxide dismutase(SOD), malondialdehyde(MDA) and glutathione peroxidase(GSH) were detected by the kit. The activity of lactate dehydrogenase(LDH), Na+-K+-ATP and Ca2+-Mg2+-ATP were detected by the kit. Survivin, Caspase-3 and Caspase-9 were detected by Western blot. Results Compared with model group, healthy controls and extraneous imperatorin 50, 100 mg/kg and nim horizon group armed into a significant rise in number of incubation period(P<0.05), the extraneous arms, starting arm into the number, the other arm into number significantly reduced(P<0.05), significantly improved pathological damage degree and the apoptosis positive cells were significantly reduced, Survivin protein level significantly increased(P<0.05), Caspase 3, Caspase-9 proteins significantly reduced(P<0.05), SOD and GSH contents significantly increased(P<0.05), MDA content significantly decreased(P<0.05), LDH activity significantly decreased(P<0.05), Na+-K+-ATP, Ca2+-Mg2+-ATP activity significantly increased(P<0.05). Conclusion Imperatorin has a protective effect on cerebral ischemia and reperfusion in rats.
