摘  要:目的探讨生长抑制因子4(ING4)基因甲基化与宫颈癌发生的关系及其临床意义。方法收集确诊的80例宫颈癌患者组织标本,并以同期的子宫良性病变行子宫全切患者的50例宫颈组织标本作为对照组。采用甲基化特异性PCR(MSP)技术检测子宫颈癌组织及对照组宫颈组织中ING4基因启动子的甲基化状态,并使用实时定量逆转录酶-聚合酶链反应(qRT-PCR)检测了ING4基因的mRNA表达水平,分析ING4基因启动子的甲基化状态与宫颈癌临床病理特征的关系。结果宫颈癌组织中ING4基因的甲基化率为82.5%(66/80),高于对照组12%(6/50)(P<0.01)。宫颈癌组织中ING4的mRNA表达(0.41±0.07)低于对照组中ING4的mRNA表达(0.87±0.16)。此外,ING4启动子区甲基化的宫颈癌组织中ING4 mRNA水平(0.37±0.06,n=66)低于ING4启动子区未甲基化宫颈癌患者组织中ING4 mRNA水平(0.46±0.11,n=14)。宫颈癌组织中ING4基因启动子区甲基化与年龄、淋巴结状态、宫颈癌的病理学分类和临床分期等变量差异无统计学意义(P>0.05)。结论ING4基因启动子区的高甲基化可能与宫颈癌的发生发展有关。Objective To investigate the relationship between the methylation of growth inhibitory factor 4(ING4) gene and cervical carcinogenesis and its clinical significance. Methods Tissue specimens of 80 patients with cervical cancer diagnosed were collected, and 50 cervical tissue specimens from patients with benign uterine lesions undergoing total hysterectomy during the same period were used as the control group. Methylation-specific PCR(MSP) technique was used to detect the methylation status of the ING4 gene promoter in cervical cancer tissues and cervical tissues of the control group, and the mRNA expression level of ING4 gene was detected by real-time quantitative reverse transcriptase-polymerase chain reaction(qRT-PCR). The expression level of ING4 mRNA was analyzed, and the relationship between the methylation status of ING4 gene promoter and the clinical case characteristics of cervical cancer was analyzed. Results The methylation rate of ING4 gene in tissue of cervical cancer(66/80, 82.5%) was significantly higher than that of control group(6/50, 12%)(P<0.01). The mRNA expression of ING4 in cervical cancer tissue(0.41±0.07) was lower than that in control group(0.87±0.16). In addition, the mRNA level in cervical cancer tissues methylated by the ING4 promoter region(0.37±0.06, n = 66) was lower than that in tissues of cervical cancer patients with unmethylated ING4 promoter region(0.46±0.11, n=14). There was no statistically significant difference between the methylation of ING4 gene promoter region and age, lymph node status, pathological classification and clinical stage of cervical cancer in cervical cancer tissues(P>0.05). Conclusion Hypermethylation of the ING4 gene promoter region may be associated with the occurrence and development of cervical cancer.
