摘  要:抗菌肽LHH1是一种来源于干酪乳杆菌的新型抗菌肽,具有良好的抗菌和抗肿瘤活性。以大肠杆菌BL21(DE3)为宿主细胞,pE-SUMO为表达载体,融合表达了抗菌肽LHH1。表达产物经过亲和色谱纯化、凝胶脱盐、肠激酶水解和RP-HPLC分离纯化后,得到了高纯度的LHH1。LHH1对金黄色葡萄球菌具有良好的抗菌活性(MIC=4 μmol/L),对山羊血红细胞具有很低的溶血毒性。此外,LHH1能够增加小鼠肠道中益生菌的丰度,并降低致病菌的丰度,发挥了改善小鼠肠道微生态的作用。研究结果可为新型抗菌肽LHH1的生产与应用提供一定的理论依据。LHH1 is a novel antimicrobial peptide that derived from Lactobacillus casei HZ-1,which possesses high antimicrobial activities and antitumor activities.In this study,pE-SUMO vector was used to express LHH1 in BL21(DE3) cell to improve the expression efficiency.The fusion protein was harvested by Ni-affinity column,desalted by gel chromatography,hydrolyzed by enterokinase and purified by RP-HPLC.The purified peptide of LHH1 exhibited an excellent antibacterial activity against Staphylococcus aureus with a minimal inhibitory concentration (MIC) value of 4 μmol/L and showed a very low hemolysis toxicity on goat blood red cell.Meanwhile,LHH1 can improve the composition of gut microflora by increasing the abundance of the probiotics and decreasing the abundance of pathogenic bacteria in the gut of C57BL/6J mice.This study could provide some theoretical basis for prodution and application of LHH1.
