摘  要:为了研究脱氧雪腐镰刀菌烯醇(doxynivalenol,DON)对蛋鸡肝脏组织形态学的影响,随机选取26周龄海兰蛋鸡120只,分为6组,每组20只鸡,试验组按体重分别灌胃1、5、10、20和50 mg/kg DON,对照组灌胃0.2 mL等量的生理盐水,试验持续6周;试验结束,分离蛋鸡肝脏,称重后做组织病理切片。结果显示:1 mg/kg组可见小叶间静脉周围炎性细胞浸润,1区肝细胞空泡化变性;5 mg/kg组可见中央静脉周围出现大量炎性细胞浸润;10 mg/kg组肝脏指数显著升高(P<0.01),伴随汇管区出现大量炎性细胞浸润,炎症病灶外围形成纤维化,小叶间静脉扩张;20 mg/kg时,肝血窦腔淤血增大,汇管周围出现局灶性溶解性坏死;50 mg/kg组肝脏指数较对照组显著降低(P<0.05),伴随汇管周围出现大量局灶性溶解性坏死,有空洞形成。由此可见,DON导致蛋鸡肝脏损伤具有剂量依赖性,低剂量呈现炎性细胞浸润,高剂量出现坏死及空洞,其中10和50 mg/kg组肝脏组织形态变化还伴随肝脏指数的升高或降低。This study was conducted to investigate the effect of deoxynivalenol(DON)on hepatic morphology in laying hens.Laying hens(n=120)were obtained and randomly allotted to 6 groups with 20 in each group.The hens in the experimental groups were exposed to 1,5,10,20 and 50 mg/kg body weight DON,respectively,every day,by intragastric administration,for 6 weeks.The hens of the control group were fed with the equivalent dosage of normal saline in the same protocol.At the end of the experiment,the liver samples were collected from the hens,were weighed,and made into histological slices.The results were that,in the 1 mg/kg DON group,there was inflammatory cell infiltration around the interlobular vein,and vacuolar degeneration of hepatocyte was also observed in Zone 1;a large number of inflammatory cells infiltrated into the centrilobular areas in the 5 mg/kg DON group.Compared with the control group,the ratio of liver weight/body weight was significantly increased in the 10 mg/kg DON group(P<0.01),which was associated with the numerical inflammatory cells,congestion and dilatation of vessels,and fibrosis in the portal tract.In addition,hepatic sinuses were congested and dilatated in the 20 mg/kg DON group,and the focal lytic necrosis was also shown in their portal tract.And,the ratio of liver weight/body weight in the 50 mg/kg DON group was remarkably decreased,compared with the control group(P<0.05).Serious focal lytic necrosis was observed in the portal tract,with a cavity present there.In conclusion,the hepatic injury of the laying hens was caused by DON in a dose-dependent manner,with infiltration of inflammatory cells at low dosage and the necrosis and cavity formation at high dosage.The lesion of hepatic histology was in line with the increase and decrease in the ratios of liver weight/body weight in the 10 and 50 mg/kg DON-treated groups,respectively.
