摘  要:目的分析限食后重饲大鼠骨密度变化,并探讨白藜芦醇对限食后重饲大鼠骨密度的影响及可能机制。方法选取SD大鼠64只并随机分为正常饮食组16只、限食后重饲组24只、限食后重饲+白藜芦醇干预组24只。再将正常饮食组分NC4组和NC12组各8只,整个实验过程均给予正常饮食;限食后重饲组分为R4组(限食4周)、RN组(限食4周后给予正常饮食8周)、RH组(限食4周后给予高脂饮食8周)各8只;限食后重饲+白藜芦醇干预组分为R4R组(限食4周的同时给予白藜芦醇干预)、RNR组(限食4周后给予正常饮食加白藜芦醇8周干预)、RHR组(限食4周后给予高脂饮食加白藜芦醇干预8周)各8只。实验结束后,检测各组大鼠体重、全身骨密度、高胰岛素-正糖钳夹60~120 min内的葡萄糖输注率(GIR 60-120),以及血清游离脂肪酸(FFA)、肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)水平。结果(1)NC4组、R4组、R4R组的体重、骨密度依次降低,NC4组、R4R组、R4组的血清FFA水平依次升高,R4组、R4R组的血清TNF-α和IL-6水平均低于NC4组(均P<0.05)。(2)RN组、RH组体重均低于NC12组,RH组骨密度低于其他两组,NC12组、RN组、RH组GIR 60-120依次降低,而血清FFA、TNF-α、IL-6水平依次升高(均P<0.05)。(3)NC12组、RN组、RNR组体重依次降低,RNR组骨密度高于其他两组且GIR 60-120高于NC12组(P<0.05);NC12组、RNR组、RN组血清FFA、TNF-α水平依次升高,且RN组血清IL-6水平高于其他两组(均P<0.05)。(4)NC12组、RH组、RHR组的体重依次降低,RH组、NC12组、RHR组的骨密度依次升高,NC12组、RHR组、RH组的GIR 60-120依次降低而FFA、TNF-α、IL-6水平依次升高(均P<0.05)。(5)相关分析结果显示全身骨密度与体重、GIR 60-120呈正相关,与FFA、TNF-α、IL-6呈负相关(均P<0.05)。逐步回归分析结果显示GIR 60-120、FFA水平是影响全身骨密度的因素(均P<0.05)。结论与限食后正常饮食重饲大鼠相比,高脂饮食重Objective To analyze bone mineral density(BMD)variations in rats refeeding after calorie restriction,and to explore the effect of resveratrol on BMD in rats refeeding after calorie restriction as well as its possible mechanism.Methods Sixty-four SD rats were enrolled and randomly divided into normal diet group(16 rats),post-caloric restriction refeeding group(24 rats),and post-caloric restriction refeeding+resveratrol intervention group(24 rats).The normal diet group was subdivided into NC4 group(n=8)and NC12 group(n=8),receiving normal diet during the experiment;the post-caloric restriction refeeding group was subdivided into R4 group(n=8)with a 4-week caloric restriction,RN group(n=8)with a 4-week caloric restriction followed by a 8-week normal diet,and RH group(n=8)with a 4-week caloric restriction followed by a 8-week high-fat diet;the post-caloric restriction refeeding+resveratrol intervention group was subdivided into R4R group(n=8)with a 4-week caloric restriction plus concurrent resveratrol intervention,RNR group(n=8)with a 4-week caloric restriction followed by a 8-week normal diet plus resveratrol intervention,and RHR group(n=8)with a 4-week caloric restriction followed by a 8-week high-fat diet plus resveratrol intervention.After the experiment ended,body weight,whole-body BMD,glucose infusion rate during 60 to 120 minutes(GIR 60-120)of hyperinsulinemic-euglycemic clamping,as well as serum levels of free fatty acid(FFA),tumor necrosis factorα(TNF-α)and interleukin 6(IL-6),were detected in rats of each group.Results(1)Body weight and BMD decreased in turn in the NC4,R4 and R4R groups,serum FFA level increased in turn in the NC4,R4R and R4 groups,the R4 and R4R groups had lower serum TNF-αand IL-6 levels than the NC4 group(all P<0.05).(2)The RN and RH groups exhibited lower body weight than the NC12 group,the RH group reported lower BMD than the other two groups,GIR 60-120 decreased whereas serum FFA,TNF-αand IL-6 levels increased in the order of the NC12 group,the RN group and the RH group(all P<0.0
