摘 要:目的:研究β-榄香烯对非小细胞肺癌细胞凋亡及氧化应激的影响,并初步探讨其可能的作用机制。方法:取对数生长期的肺腺癌A549细胞,分为未处理对照组和低、中、高剂量(10、25和50μg/mL)β-榄香烯处理组。采用AnnexinⅤ/碘化丙啶(propidium iodide,PI)双染色法检测细胞的凋亡率;DCFH-DA荧光探针法检测细胞内活性氧(reactive oxygen species,ROS)水平;生化试剂盒检测细胞内超氧化物歧化酶(superoxide dismutase,SOD)的活性、总谷胱甘肽(total glutathione,T-GSH)和丙二醛(malondialdehyde,MDA)的含量以及总抗氧化能力(total antioxidant capacity,T-AOC);实时荧光定量PCR及蛋白质印迹法检测核因子E2相关因子2(nuclear factor E2-related factor 2,Nrf2)和血红素加氧酶-1(heme oxygenase-1,HO-1)mRNA及蛋白的表达水平。结果:与对照组相比,β-榄香烯处理后A549细胞凋亡率均明显提高(P值均<0.01);细胞内ROS水平上升,SOD活性、T-GSH含量和T-AOC均明显降低(P值均<0.01),MDA的含量明显升高(P<0.01),Nrf2和HO-1 mRNA及蛋白表达水平均显著降低(P值均<0.01);其中,高剂量β-榄香烯处理组的作用效果最显著(P<0.01)。结论:β-榄香烯对非小细胞肺癌细胞的凋亡具有促进作用,其机制可能与下调Nrf2/HO-1通路和促进氧化应激有关。Objective:To investigate the effects ofβ-elemene on the apoptosis and oxidative stress of non-small cell lung cancer cells,and to explore the possible mechanism.Methods:Lung adenocarcinoma A549 cells in logarithmic growth phase were divided into the blank control group and the test groups treated with low,medium and high dose(10,25,50μg/mL)β-elemene separately.Apoptosis was detected by AnnexinⅤ/PI double staining method,and the level of reactive oxygen species(ROS)in the cells were detected by DHFH-DA staining method.Intracellular superoxide dismutase(SOD)activity,total glutathione(T-GSH)and malondialdehyde(MDA)content,and total antioxidant capacity(T-AOC)were detected by biochemical kit.The mRNA and protein expression levels of nuclear factor E2-related factor 2(Nrf2)and heme oxygenase-1(HO-1)were detected by real-time fluorescent quantitative PCR and Western blotting,respectively.Results:Compared with the blank control group,the apoptosis rate of A549 cells increased significantly afterβ-elemene treatment(P<0.01),the intracellular ROS level notably increased,SOD activity,T-GSH content and T-AOC decreased significantly(all P<0.01),and MDA content increased significantly(P<0.01).The mRNA and protein expression levels of Nrf2 and HO-1 were significantly decreased(all P<0.01).Moreover,the treatment with high dose ofβ-elemene was the most significant(P<0.01).Conclusion:β-Elemene promoted the apoptosis of non-small-cell lung cancer cells,and the mechanism may be related to the down-regulation of Nrf2/HO-1 axis and the promotion of oxidative stress reaction.
