摘  要:目的:探索甲基萘醌-4(MK-4)在少突胶质细胞缺氧环境中的作用。方法:建立少突胶质细胞缺氧模型,并给予MK-4,CCK-8检测细胞活性变化;二氢乙锭染色检测细胞活性氧(ROS)释放情况;qRT-PCR检测炎症因子白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)及生长停滞特异性蛋白6(Gas6)mRNA表达变化。结果:MK-4可显著提高缺氧少突胶质细胞的活性,减少ROS的释放,下调IL-1β、IL-6、TNF-α等炎症因子的表达,促进Gas6的表达。结论:MK-4对少突胶质细胞缺氧性损伤具有保护作用。Objective:To investigate the effect of menaquinone-4(MK-4)on oligodendrocytes under hypoxic condition.Methods:The CCK-8 assay was used to determine the effect of MK-4 on cell viability in hypoxic conditions of oligodendrocytes.Dihydroethidium staining was performed to detect the release of reactive oxygen species(ROS).The mRNA expression changes of cytokines interleukin-1β(IL-1β),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α)and growth arrest specific-6(Gas6)were detected by real time RT-PCR.Results:MK-4 could greatly increase the cell viability of oligodendrocytes and decrease the release of ROS after hypoxic injury.Meanwhile,the mRNA expression levels of IL-1β,IL-6,TNF-αand Gas6 showed significant downregulation.MK-4 could promote the expression of Gas6.Conclusion:MK-4 has a protective effect on oligodendrocytes during hypoxic injury.
