摘 要:目的探讨结肠癌发生机制中内质网氧化硝化应激及细胞色素P450(CYP450)表达调控机制。方法将64只小鼠随机分为病毒组和对照组,每组32只。病毒组小鼠进行腹腔接种小鼠巨细胞病毒(MCMV)悬液20μL[半数组织培养感染量(TCID50)=107.21/mL)]建立MCMV肠道模型;对照组小鼠进行腹腔注射等量生理盐水。分别于第3、7、14和28天处死小鼠,采用RT-PCR法检测小鼠肠道细胞内CYP450 mRNA水平,采用ELISA法检测小鼠血清ALT水平,采用免疫组化法检测小鼠肠道细胞内转录因子C/EBP同源蛋白(CHOP)表达水平及分布情况。结果成功建立小鼠MCMV模型。病毒组在各时间点的ALT和CYP450表达水平均高于对照组(P<0.05)。结论内质网应激相关分子CYP450在小鼠MCMV肠道模型中的表达水平均明显提高;CYP450在人结肠癌细胞中的高表达可增加对内质网氧化硝化应激,从而促进细胞增殖。Objective To investigate the endoplasmic reticulum oxidative nitrification stress and cytochrome P450 expression regulation in the pathogenesis of colon cancer.Methods Sixty-four mice were randomly assigned into two groups with 32 mice each.The mouse models with cytomegalovirus intestinal in group A were established by intraperitoneally injection of murine cytomegalovirus(MCMV)virus suspension 20μL(TCID50=107.21/mL).The mice of group B were intraperitoneally injected the same amount of normal saline as the controls.The mice were executed and sample were taken at different points of the 3rd,7th,14th and 28th day after model establishment for the detection of cytochrome P450 mRNA by RT-PCR.The level of alanine transaminase(ALT)in serum and the expression of homologous protein C/EBP transcription factors(CHOP)in intestinal cells of mice were detected by ELISA and immunohistochemical method,respectively.Results The mouse cytomegalovirus intestinal model was successfully established.Serum ALT level,the CHOP mRNA and cytochrome P450 protein expressions were significantly higher in group A than those in group B on the 3rd,7th,14th and 28th day after model establishment.Conclusion The endoplasmic reticulum stress-related molecule cytochrome P450 in the intestinal tract of MCMV mice is increased significantly.The high expression of cytochrome P450 in human colon cancer cells could increase the oxidative nitrification stress on endoplasmic reticulum and promote the cell proliferation.
