摘 要:目的探讨微小RNA-6775-3p(miR-6775-3p)在乳腺癌组织中的甲基化状态及其临床意义。方法甲基化特异性PCR检测63例乳腺癌患者乳腺癌组织及其癌旁正常组织中miR-6775-3p启动子区CpG岛甲基化水平。实时荧光定量PCR法分析乳腺癌组织中miR-6775-3p表达。分析miR-6775-3p启动子区甲基化状态与乳腺癌患者临床病理参数及预后关系。结果乳腺癌组织中miR-6775-3p启动子区甲基化阳性率高于癌旁正常组织(76.2%vs.19.0%)(P<0.01)。miR-6775-3p启动子区甲基化阳性率与TNM分期、肿瘤浸润深度、淋巴结转移及远处转移或复发密切相关(P<0.05)。与15例miR-6775-3p启动子区未甲基化患者比较,48例miR-6775-3p启动子区甲基化患者乳腺癌组织中miR-6775-3p基因表达水平较低(2.743±0.257 vs.4.521±0.399)(P<0.01),生存时间较短[(40.44±4.18)个月vs.(48.80±2.59)个月](P<0.05),累计生存率较低(75.0%vs.80.0%)(P<0.05)。结论miR-6775-3p甲基化可能导致其在乳腺癌组织中低表达,可能与乳腺癌的肿瘤浸润、转移及预后相关。Objective To explore the methylation status of miR-6775-3 p in breast cancer tissues and its clinical significance.Methods The methylation level of CpG island methylation in the promoter region of miR-6775-3 p in breast cancer tissues(group A)and tumor-adjacent normal tissues(group B)in 63 patients with breast cancer were detected by methylation specific PCR.The miR-6775-3 p expression in group A was detected by quantitative real-time PCR.The relationship between the methylation status of miR-6775-3 p promoter region and clinicopathological parameters and prognosis in group A was analyzed.Results The methylation positive rate of the promoter region of miR-6775-3 p in group A was higher than that in group B(76.2%vs.19.0%)(P<0.01),which was closely related to TNM stage,tumor infiltration depth,lymph node and distant metastasis or recurrence(P<0.05).Compared to the patients(15 cases)without the promoter of miR-6775-3 p methylation,the patients(48 cases)with the promoter of miR-6775-3 p methylation had a lower level of miR-6775-3 p gene expression in breast cancer(2.743±0.257 vs.4.521±0.399)(P<0.01),shorter survival time[(40.44±4.18)months vs.(48.80±2.59)months](P<0.05),and lower cumulative survival rate(75.0%vs.80.0%)(P<0.05).Conclusion Methylation in the promoter region of miR-6775-3 p gene may cause its low expression in breast cancer tissues,which may be related to tumor invasion,metastasis and prognosis of the patients with breast cancer.
