摘  要:目的采用多指标正交试验优选黄芪女贞方菌质的最佳醇沉工艺。方法以黄芪甲苷、特女贞苷转移率及干膏得率为评价指标,选择药液浓缩比(A)、乙醇浓度(B)、含醇量(C)、醇沉时间(D)为考察因素,进行L9(34)正交试验,结合层次分析法及多指标试验全概率评分法优选最佳醇沉工艺。结果建立了黄芪女贞方菌质中黄芪甲苷、特女贞苷含量的HPLC测定方法。以黄芪甲苷转移率为评价指标,影响因素为C3>B3>A2>D2;以特女贞苷转移率为评价指标,影响因素为A3>D2>C2>B3;以干膏得率为评价指标,影响因素为A3>C1>B3>D2。加权评分优化后确定的最佳醇沉工艺为:菌质∶提取液=1∶1.5,采用85%乙醇,含醇量75%,醇沉18h。结论黄芪女贞方菌质中黄芪甲苷和特女贞苷在优选的醇沉工艺条件下转移率较高,达到较好的纯化效果,工艺稳定可行,可为进一步研究提供依据。Objective To optimize the alcohol precipitation technology of mycoplasm from Huangqi Nvzhen Prescription through multi-index orthogonal experiment.Methods The transfer rates of astragalosides and specnuezhenide and the yield of dry paste were set as evaluation indexes;The ratio of concentration of liquid medicine(A),ethanol concentration(B),alcohol content(C)and alcohol precipitation time(D)were selected as the study factors;the L9(34)orthogonal test was carried out.The optimal alcohol precipitation process was preferred by the method of analytic hierarchy process and the multi-index trial full-probability scoring method.Results A method for the determination of astragalosides and specnuezhenide in mycoplasm from Huangqi Nvzhen Prescription by HPLC was established.With the transfer rate of astragalosides as evaluation index,the influencing factors were C3>B3>A2>D2;with the transfer rate of specnuezhenide as evaluation index,the influencing factors were A3>D2>C2>B3,and the yield of dry paste was A3>C1>B3>D2.The optimal alcohol precipitation process after the optimization of the weighted score were:the concentration ratio of the liquid medicine(mycoplasm:extracting solution)was 1:1.5;the ethanol concentration was 85%;the alcohol content was 75%;the alcohol precipitation time was 18 h.Conclusion The transfer rates of astragalosides and specnuezhenide in the optimal alcohol precipitation process are high,and better purification effect is achieved.The process is stable and feasible,which can provide basis for further study.
