摘  要:为了探索小麦抗叶锈病基因Lr19抗叶锈病的分子机理,并筛选参与其抗病性防御反应的相关基因,本研究对接种小麦叶锈菌生理小种PHNT及其EMS突变体后的小麦材料‘TcLr19’进行转录组测序及分析。与亲和互作反应相对比,在非亲和互作反应中筛选到2114个差异表达基因,其中上调差异表达基因1189个,下调差异表达基因925个。对1189个上调差异表达基因进行GO功能分析,部分差异表达基因被注释为在真菌的防御反应中发挥抗病功能;KEGG分析表明,一些与抗病相关的差异表达基因被富集到了过氧化物酶、氨基糖/核苷酸糖代谢和角质生物合成信号通路。同时,发现一些与Lr19同在7D染色体上的基因受叶锈菌诱导表达,并在非亲和互作反应中特异性表达。总之,本研究初步明确了参与Lr19抗叶锈病防御反应过程中抗病相关基因的种类以及抗病代谢通路,为探索Lr19的抗叶锈病机制提供了理论基础。To understand the molecular mechanism of wheat leaf rust resistance gene Lr19 under the stress of leaf rust pathogen,and to identify genes closely related to resistance to leaf rust pathogen,transcriptome sequencing was conducted on TcLr19 inoculated with Puccinia triticina(Pt)variety PHNT and its EMS mutants.A total of 2114 differentially expressed genes(DEGs)were obtained in incompatible combination compared with compatible combination,of which 1,189 were up-regulated and 925 were down-regulated.1189 up-regulated DEGs were subjected to GO functional classification and KEGG enrichment analysis,and some of DEGs were annotated as proteins related to wheat defense to Pt.KEGG analysis showed that some DEGs related to disease resistance were enriched to peroxidation lysosomal signaling pathway.In addition,we found that some genes located on the 7 D chromosome,which was consistent with Lr19,were expressed differently in incompatible combination.All the above results preliminarily clarified the resistance-related genes to Pt and disease-resistant metabolic pathways involved in the reaction of Lr19 against Pt,which provided a theoretical basis for exploring resistant mechanism of Lr19.
