摘  要:为满足快速高效评价微生物培养基生长率的需求,提出一种新的微生物标准物质研制思路,并研制出具有2种特性量值的大肠杆菌标准物质。研制过程中同时使用平板计数琼脂(plate count agar,PCA)培养基和结晶紫中性红胆盐琼脂(violet red bile agar,VRBA)培养基进行均匀性评估、稳定性评估、6家实验室合作定值以及不确定度评定。实验结果表明:大肠杆菌标准物质的均匀性和稳定性良好,可以在-20℃下稳定保存6个月。该标准物质基于PCA培养基的标准值为(7.1±1.1)×10^3 CFU/g,基于VRBA培养基的标准值为(6.1±1.0)×10^3 CFU/g。使用该标准物质对多个PCA培养基和VRBA培养基的生长率进行评价,评价结果和国标方法有很好的相关性。综上所述,研制的大肠杆菌标准物质可以用于多种培养基的生长率评价,具有方便快捷的优点。In order to meet the requirements of the productivity ratio evaluation of the microbiological media quickly and efficiently,a new idea for the development of microbial reference materials was proposed and a reference material of Escherichia coli(E.coli)with 2 characteristic values was developed.The plate count agar(PCA)and violet red bile agar(VRBA)were used together during the homogeneity and stability evaluation,the value determination,and the uncertainty evaluation.The result indicated that the reference material of E.coli bearing excellent homogeneity and stability could be stored for six months at-20℃.The certified values were(7.1±1.1)×10^3 CFU/g based on PCA and(6.1±1.0)×10^3 CFU/g based on VRBA.In addition,the productivity ratio of several brands PCA and VRBA was evaluated based on the reference material,and the results had a good correlation with that of the method in the national standard.In conclusion,the developed reference material of E.coli was valid for productivity ratio evaluation of various microbiological medium,which could be more convenient and efficient.
