摘 要:目的联合检测DLC-1和FAK基因在正常结肠黏膜、结肠腺瘤及结肠癌组织中的表达水平,探讨两者在结肠黏膜癌变过程中的相互关系及临床意义。方法选择2017年12月至2018年10月就诊于内蒙古包钢医院消化内科的患者,收集对其进行结肠镜检查或治疗时获取的结肠组织标本,根据结肠镜下形态学观察及病理诊断分为正常黏膜组、结肠腺瘤组及结肠癌组。采用实时荧光定量聚合酶链反应(RT-PCR)法检测DLC-1 mRNA和FAK mRNA在3种组织中的表达水平。结果DLC-1 mRNA在正常黏膜组、腺瘤组及结肠癌组中的相对表达量分别为0.889±0.090、0.614±0.071、0.216±0.059,呈逐渐降低趋势,差异有统计学意义(P<0.001)。FAK mRNA在正常黏膜组、腺瘤组及结肠癌组中的相对表达量分别为0.674±0.219、1.329±0.375、3.136±0.454,呈逐渐升高趋势,差异有统计学意义(P<0.001)。各组的DLC-1 mRNA、FAK mRNA表达均呈负相关。结论DLC-1 mRNA与FAK mRNA的表达在结肠腺瘤癌变过程中的不同阶段存在差异性,且两者呈负相关。联合检测DLC-1 mRNA与FAK mRNA的表达可能有助于结肠癌的早期诊断。Objective This paper intends to jointly detect the expression of DLC-1 and FAK genes in normal colon mucosa,colon adenoma,and colon cancer tissues,and explore the relationship and clinical significance of the two in the process of colon mucosal carcinogenesis.Methods Patients who were treated in the Gastroenterology Department of Inner Mongolia Medical Baogang Hospital from December 2017 to October 2018 were selected and assigned into the normal mucosal group,the colon adenoma group,and the colon cancer group based on the colon tissue specimens obtained during colonoscopy or treatment according to colonoscopy morphological observation and pathological diagnosis.Real-time fluorescent quantitative polymerase chain reaction(RT-PCR)was used to detect the expressions of DLC-1 mRNA and FAK mRNA in the three tissues.Results The relative expression of DLC-1 mRNA in the normal mucosa group,the adenoma group,and the colon cancer group were 0.889±0.090,0.614±0.071,and 0.216±0.059,respectively,showing a gradual decrease,with a statistically significant difference(P<0.001).The relative expression of FAK mRNA in the normal mucosa group,the adenoma group,and the colon cancer group were 0.674±0.219,1.329±0.375,and 3.136±0.454,respectively,showing a gradual increase,with a statistically significant difference(P<0.001).The expressions of DLC-1 mRNA and FAK mRNA in each group were negatively correlated.Conclusions The expressions of DLC-1 mRNA and FAK mRNA are different in different stages of colon adenoma carcinogenesis,and the two are negatively correlated.The combined detection of the expressions of DLC-1 mRNA and FAK mRNA may be helpful for the early diagnosis of colon cancer.
