茶树精油对变异链球菌抑菌浓度及效力的探究

摘　　要：目的:探讨茶树精油(TTO)对变异链球菌(S.mutans)体外抑菌的作用。方法:利用牛津杯法检测TTO对S.mutans的抑菌效果;标准微量肉汤稀释法测定TTO对S.mutans的最小抑菌浓度(MIC)和最小杀菌浓度(MBC),MTT染色法计算不同时间下TTO的抑菌率和杀菌时间,评估TTO对S.mutans抑菌效力。对照组分别为0.2%氯己定(CHX)和0.5%Tween-80。结果:实验结果显示抑菌环(SBR)大小为(11.36±1.68)mm^(20.26±3.64)mm;MIC为0.125%,MBC为0.25%。0.25%TTO在24 h内可以完全杀灭细菌,0.125%TTO在12、24内抑菌率分别为(90.97±0.77)%、(69.94±4.46)%,与对照组相比,差异无统计学意义(t 12=7.390,P 12=0.947;t 24=4,627,P 24=0.219)。0.25%TTO在48 h时抑菌率下降为68.63%±0.47%,与对照组相比差异无统计学意义(t=1.715,P=0.068)。结论:TTO对S.mutans抑菌作用显著,0.125%TTO可抑制S.mutans活性,0.25%TTO可起到灭菌作用,抑菌效果稳定。Objective:To study the effects of tea tree oil(TTO)against Streptococcus mutans(S.mutans).Methods:The antimicrobial activity of TTO on S.mutans was tested by agar disk diffusion method,the minimal inhibitory concentration(MIC)and minimal bactericidal concentration(MBC)were detected by Micro broth dilution method.The bacteriostatic rate of different concentrations of TTO was detected by colony counting method.The bactericidal activity of TTO against S.mutans was tested by MTT.0.2%chlorhexidine and 0.5%Tween-80 were used as the controls.Results:The size of becteriostatic ring(mm)for S.mutans were in the range of 12.4±0.62~4±0.43.The MIC and MBC of TTO were 0.125%and 0.25%.In the 0.25%TTO group,the bacteria were completely killed at 24 hours.The bacteriostatic rates of 0.125%TTO within 12 and 24 hours were 90.97%±0.77%and 69.94%±4.46%respectively and had no significantly difference competed with the control groups(t 12=7.390,P 12=0.947;t 24=4,627,P 24=0.219).The bacteriostatic rate of 0.25%TTO was 68.63%±0.47%for 48 hours and had no significant difference with the control groups(t=1.715,P=0.068).Conclusion:TTO has a significant bacteriostatic effect on S.mutans.0.125%TTO can stably inhibit S.mutans and 0.25%TTO can sterilize the bacteria.
