摘  要:目的:研究实验性牙移动造成的咬合干扰引起大鼠咬肌损伤的程度及骨骼肌损伤标志物的表达变化。方法:54只SD大鼠分为3组(n=18)。对照组(Con)大鼠无干预,咬合干扰(Exp)大鼠以正畸皮圈推大鼠左侧上颌及右侧下颌第三磨牙向远中移动,咬合干扰加EDTA(Exp+EDTA)组大鼠加注EDTA-Na 2,90 mg/kg,1次/d。此后1、2、4周观察咬肌组织学及超微结构改变,检测咬肌中骨骼肌肌钙蛋白抑制亚基(sTnI)、结蛋白(desmin)的表达,及血清中肌酸激酶(CK)、乳酸脱氢酶(LDH)和肌红蛋白(Mb)含量。结果:咬合干扰组大鼠咬肌组织学形态及其他骨骼肌损伤标志物没有明显变化,但出现线粒体肿胀等超微结构改变,线粒体内Ca^2+含量明显增加,并且在阻断Ca^2+后可明显缓解超微结构损伤。结论:实验性牙移动形成的咬合干扰造成大鼠咬肌轻微损伤,表现为超微结构变化,且与细胞内Ca^2+稳态失调有关。Objective:To investigate the injury of masseter induced by occlusal interference via experimental tooth movement and the expression of skeletal muscle injury markers in rats.Methods:Maxillary left and mandibular right third molars of 36 rats were moved distally by the propulsive force of an elastic band,18 of them were used as the experimental group(Exp group)and 18 were given EDTA-Na 2 injection in masseter 90 mg/kg,1/d(Exp+EDTA group).18 rats without treatment were used as the controls(Con group).1,2 and 4 weeks after operation.The histological and ultrastructural alterations of masseter were observed by HE staining and TEM.The expression of skeletal troponin inhibitory subunit(sTnI)and desmin in masseter tissue,the expression of serum creatine kinase(CK),lactate dehydrogenase(LDH)and myoglobin(Mb)were detected.Results:In the Exp groups there were no significant changes in masseter histology and other injury markers,but there were ultrastructural injuries such as morphological alterations of mitochondrial,the content of Ca^2+in the mitochondria increased significantly.However,ultrastructural changes could be alleviated after blocking Ca^2+in Exp+EDTA groups.Conclusion:Occlusal interference induced by experimental tooth movement can cause minor injury to the masseter in rats,which is characterized by ultrastructural changes and is associated with intracellular Ca^2+homeostasis disorder.
