摘 要:目的研究黄芩素与γ-环糊精(γ-CD)形成包合物的制备过程和结构表征,不同pH值对包合物的影响,包合机制及其抗氧化活性。方法紫外-可见吸收光谱法、荧光光谱法和核磁共振法(NMR)研究黄芩素与γ-CD在溶液中形成的包合物;加热回流共沉淀法制备黄芩素与γ-CD的固体包合物;红外光谱法(IR)对形成的包合物进行表征,荧光光谱法测定包合物的包合常数(K)及包合比(n);双倒数曲线法研究包合前后对自由基1,1-二苯基-2-三硝基苯肼(DPPH·)的清除速率。结果在不同的pH值溶液中,γ-CD对黄芩素具有不同的包合能力,γ-CD最适合在中性介质中与黄芩素形成包合物,在实验浓度范围内,按1∶1形成包合物,包合比为1067,包合物对自由基DPPH·的清除活性更强;NMR确定包合物的结构。结论在实验条件下,黄芩素分子从γ-CD大口端进入到空腔内形成稳定的包合物,且抗氧化活性增加。OBJECTIVE To study the preparation,structure,inclusion mechanism and antioxidant activity ofγ-cyclodextrin(γ-CD)inclusion complex of baicalein.METHODS The inclusion complex of baicalein andγ-CD in solution was studied by ultravioletvisible absorption spectroscopy,fluorescence spectroscopy and nuclear magnetic resonance(NMR).The solid inclusion complex of baicalein andγ-CD was prepared by coprecipitation method.The inclusion complex was characterized by infrared spectroscopy(IR).The inclusion constant(K)and inclusion ratio(n)of the inclusion complex were determined.RESULTS In solutions of different p H values,γ-CD had different inclusion ability to baicalein,and neutral medium was the most suitable forγ-CD to form inclusion complex with baicalein.In the range of experimental concentration,baicalein andγ-CD formed inclusion complex at ratio of 1∶1,and the inclusion ratio was 1067.Baicalein/γ-CD inclusion complex had more stronger scavenging ability for free radical DPPH·than free baicalein.The structure of the inclusion complex was determined by NMR.CONCLUSION Under the experimental conditions,baicalein molecule is encapsulated into the cavity ofγ-CD from the large mouth,forming a stable inclusion complex,and the antioxidant activity is increased.
